摘要
对淡水区一株生产红色素的菌株进行了鉴定,并对所生产的红色素进行了分离、纯化与结构表征.菌株的鉴定采用生理生化法与16Sr DNA测序结合进行,鉴定结果表明,该菌属于粘质沙雷氏菌.利用柱色谱对湿重1.5g浓缩后的乙醇提取物进行分离和纯化,结果得到大约1.3g的红色素样品.经薄层色谱、紫外-可见、红外、质谱及核磁共振氢谱分析,鉴定该色素为灵菌红素(prodigiosin).
A red pigment-producing strain isolated from fresh water was identified. The produced pigment was subsequently separated, purified and characterized. Identification of strain was carried out using biochemical and physiological methods combined with 16SrDNA sequencing. The final results confirmed that the strain belonged to Serratia marcescens. Using column chromatography,a wet weight of 1.5g ethanol extract was separated, purified and resulted in about 1.3g red pigment product. TLC and HPLC test results showed a purity of above 95%. The red pigment was identified as prodigiosin by the methods of TLC, UVvisible, infrared spectrometry, mass spectrometry and nuclear magnetic resonance spectroscopy.
