摘要 目的:探讨不同运动负荷对大鼠血管内皮功能影响,并分析超声评估的可行性。方法:随机将24只SD雄鼠按运动量分为运动缺乏组(E0)、自由活动组(E1)及有氧训练组(E2),分别进行12周不同运动负荷干预。干预结束后超声检查,测量给药前后大鼠腹主动脉内径,在体评估血管内皮功能。采用Western blot技术检测离体腹主动脉内皮型一氧化氮合酶(e NOS)及诱导型一氧化氮合酶(i NOS)的表达情况。结果:1三组大鼠腹主动脉均未见斑块形成,E0组收缩期峰值流速比E1组明显升高(P<0.05)。2内皮依赖性舒张功能,与E1组相比,E0组降低,E2组增强(P<0.05);内皮非依赖性舒张功能,三组比较无明显差异。3e NOS蛋白表达水平,与E1组相比,E0组下降,E2组上调(P<0.05);i NOS蛋白表达水平,三组比较无明显差异。结论:缺乏运动引起大鼠血管内皮舒张功能受损,有氧训练起改善作用,超声有助于在体评估。 Objective: To investigate the effects of different exercise load on vascular endothelial function of rats, and ana- lyze the feasibility of ultrasonographic evaluation. Method: Twenty-four male rats were randomly divided into physical inactivity (E0) group, free moving (E1) group and aerobic exercise (E2) group, receiving 12-week intervention of different exercise load respectively. After that, inner diameter of abdominal aorta before and after drug administration were measured by ultrasound to evaluate vascular endothelial function in vivo. The protein expressions of endothelial nitricoxide synthase (eNOS) and inducible nitricoxide synthase (iNOS) in vitro abdominal aorta were determined by Western blot. Result: ①No plaque formation was found in abdomen aorta of three groups. Arteria peak systolic velocity of E0 was higher than that of E1 (P〈 0.05). ②Endothelium dependent dilatation(EDD), compared with El, less- ened in E0 and strengthened in E2 (P〈0.05). There was no difference in endothelium independent dilatation (EID) of three groups. ③The eNOS expressions, compared with E1 lessened in E0 and strengthened in E2 (P 〈 0.05). There was no difference in eNOS expression of three groups. Conclusion: Physical inactivity may impair vascular endothelial function of rats, while aerobic exercise im- proves it. Ultrasonographic evaluation is feasible.
