摘要
目的检测乳腺癌和癌旁正常乳腺组织中乙酰肝素酶(HPSE)mRNA和蛋白表达情况,探讨其与临床病理特征的关系。方法应用反转录聚合酶链反应法检测50例乳腺癌组织和20例癌旁正常乳腺组织中HPSE mRNA的表达,同时应用免疫组织化学法检测组织标本中HPSE蛋白的表达,并与临床病理特征进行相关性分析。结果HPSE mRNA和蛋白在乳腺癌组织中阳性表达率(70.0%、68.0%)均显著高于癌旁正常组织(25.0%、0.0%)(P<0.01)。乳腺癌组织中HPSE mRNA和蛋白的表达与肿瘤大小、组织学分化程度、TNM分期及淋巴结转移相关(P<0.05)。结论 HPSE mRNA和蛋白在乳腺癌组织中高表达,且与乳腺癌的发生、发展有关。
Objective To detect the expression of heparanase( HPSE) protein and mRNA in patients with breast cancer,and investigate the relationship between HPSE and clinical pathological characteristic. Methods Reverse transcription-polymerase chain reaction method and immunohistochemical method were used to detect the expression of HPSE mRNA and protein respectively in 50 breast cancer tissue and 20 adjacent normal breast tissue. The correlation between HPSE expression and clinical pathological characteristic was analyzed. Results The positive expression rate of HPSE mRNA and protein was70. 0% and 68. 0% in breast cancer tissue,which was significantly higher than that in adjacent tissue(25. 0%,0. 0%)( P 0. 01). HPSE mRNA expression in breast cancer was closely related with tumor size,TNM stages,histological grade and axillary lymph nodes metastasis( P 0. 05). Conclusion The expression of HPSE mRNA and protein in breast cancer tissue is high,it may be involved in the invasion and metastasis of breast cancer.
