摘要
目的:研究桃红四物汤对光老化小鼠皮肤组织中基质金属蛋白酶-1(MMP-1)、基质金属蛋白酶-3(MMP-3)mRNA及血清中肿瘤坏死因子-α(TNF-α)、白细胞介素-1(IL-1)的影响。方法:将60只小鼠随机分为6组,空白对照组、模型对照组、阳性对照组(VE组)和中药低、中、高剂量组,对模型对照组、阳性对照组(VE组)和中药低、中、高剂量组制备光老化模型,空白对照组正常饲养。空白对照组和模型对照组给生理盐水,高、中、低剂量组给相应剂量药液,阳性对照组(VE组)给予维生素E滴剂,于造模前30 min灌胃。造模完成后,采用Elisa和RT-PCR方法检测血清中TNF-α、IL-1含量和皮肤组织中MMP-1、MMP-3 mRNA含量的表达水平。结果:与空白对照组比较,模型对照组血清中TNF-α、IL-1和皮肤组织中MMP-1、MMP-3 mRNA含量的表达水平显著上升,差异具有统计学意义(P<0.01);与模型对照组组比较,中药低、中、高剂量组血清中TNF-α、IL-1和皮肤组织中MMP-1、MMP-3 mRNA含量的表达水平显著下降,差异具有统计学意义(P<0.01)。结论:桃红四物汤在皮肤光老化过程中具有保护作用。
Objective: To study the effect of on expressions of MMP-1,MMP-3 mRNA in skin tissue and TNF-α,IL-1 in blood serum in photoaged mice. Methods: 60 mice were divided into 6 groups,blank control group,model control group,positive control group,high dose group,moderate dose group and low dose group. Blank control group was normally fed,other groups were established photoaging model. Before 30 minutes of the preparation of photoaging model,we gave saline to blank control group and model control group,corresponding dose of liquid to high,medium and low dose group,vitamin E drops to positive control group. The expression levels of expressions of MMP-1,MMP-3 mRNA in skin tissue were measured by ELISA and TNF-α,IL-1 in blood serum in each group were measured by RT-PCR. Results: Compared with blank control group,expressions of MMP-1,MMP-3 mRNA in skin tissue and TNF-α,IL-1 in blood serum of model control groupwere significantly higher( P〈0. 01); compared with groupⅡ model control group,expressions of MMP-1,MMP-3 mRNA in skin tissue and TNF-α,IL-1 in blood serum of high,medium and low dose groupwere significantly lower( P〈0. 01). Conclusion: Taohongsiwu decoction has a protective effect on the skin photoaging process.
