几种不同破壁方法对提取酵母内容物的影响 被引量:3

摘要 以氨基态氮含量为指标,利用单因素的方法比较超声波法、酶法、自溶法以及高压均质法破碎酵母细胞壁的效果,探索在提取酵母细胞内容物过程中细胞破壁技术的工艺优化问题,并发现自溶法为最佳破壁手段。通过设计四因素三水平的正交试验,优化自溶法的最优工艺。结果表明,在自溶温度50℃,自溶p H值5.0,自溶时间26 h,并加入3%的Na Cl作为自溶促进剂时,其氨基态氮含量最高为0.114 g/100 m L,约占酵母干质量2.27%。 In this paper,the optimal conditions of yeast cell wall breaking is studied systematically and finally confirmed. In order to discover how cell wall breaking method including ultrasonic wave, enzymatic, autolysis and high pressure homogenization affected the process of yeast extraction, single factor analysis method is applied and analyzed with concentration of amion nitrogen as marker. Autolysis is found to be the preferable. To investigate the optimization conditions,three conditions such as temperature,p H and time are then optimized by orthogonal array design to be:50 ℃,p H 5.0 and26 hours with addition of 3% Na Cl as autolysis accelerator. Under these conditions, the highest yield of amino-nitrogen content of 0.114 g/100 m L,which accounts for about 2.27% of the dry yeast.
