摘要
目的研究双泽清脂颗粒的降血脂作用和安全性。方法采用高脂饲料(1%胆固醇、0.3%胆酸钠、10%猪油、88.7%普通饲料)喂养SD大鼠2周制备高脂血症动物模型。将高脂血症动物模型大鼠随机分为高脂对照组及双泽清脂颗粒高、中、低剂量组,持续给药6周后,与普通饲料组大鼠内眦采血分离血清,测定高脂血症模型大鼠血清中血脂4项(TC、TG、HDL-C、LDL-C)的水平,同时对双泽清脂颗粒进行急性毒理试验。结果双泽清脂颗粒低、中、高剂量组均能显著降低高脂血症模型动物血清中TG、TC、LDL-C含量,升高HDL-C含量(P<0.01);最大给药试验小鼠未出现急性毒性反应;双泽清脂颗粒对PPARγ2m RNA和C/EBPαm RNA的表达有抑制作用。结论本制剂急性毒性低,有明显的降血脂作用。
Objective To determine the hypolipidemic effect and safety of Shuangze Qingzhi granules. Methods The high cholesterol rat model was prepared which fed on high fat diet(1% cholesterol, 0.3% sodium cholate, 10%lard, 88.7% normal diet) for 2 weeks. The high cholesterol rats were randoml y divided a high-fat control group, and Shuangze Qingzhi granule high, medium and low dose groups. The administration lasted 6 weeks. The serum lipids(TC, TG, HDL-C, and LDL-C) levels were determined, while the acute toxicological test was conducted on Shuangze Qingzhi granules. The acute toxicity of Shuangze Qingzhi granules was assayed by the maximum dosage experiment. The effects of different dosages of Shuangze Qingzhi granules in the hyperlipidemia model and the possible molecular mechanism were determined. Results The maximum dosage did not show the acute toxicity; Shuangze Qingzhi granule low, midium, high dose groups significantly reduced the contents of the serum TG, TC and LDL-C of hyperlipidemia animal model, and increased the content of HDL-C(P 0.01). Shuangze Qingzhi granules inhibited the expression of PPARγ2 m RNA and C/EBPα m RNA. Conclusion The acute toxicity of this preparation is low and the hypolipidemic effect is clear.
