摘要
目的:研究外源性骨髓间充质干细胞(BMSCs)对大肠埃希菌致大鼠前列腺炎的抑制作用。方法:贴壁选择法分离、培养、扩增BMSCs。将sD大鼠50只随机分为急性细菌性前列腺炎(ABP)组、ABP+BMSCs组、慢性细菌性前列腺炎(CBP)组、CBP+BMSCs组和正常对照组,每组10只。在小动物超声引导下向大鼠前列腺两侧叶内注射大肠埃希菌建立前列腺炎症模型,注入细菌1~14d为急性炎症期,4—12周为慢性炎症期,对照组注入等量PBS。然后将BMSCs注入ABP+BMSCs组和CBP+BMSCs组大鼠前列腺内,观察移植BMSCs2周后大鼠前列腺病理学变化并做炎症评分,RT—PCR扩增前列腺组织中炎症因子IL—β、TNF—αmRNA,ELISA检测IL-1β、TNF-α蛋白含量,统计分析各组表达差异。结果:前列腺组织病理学提示炎症组前列腺组织呈典型炎症病理变化,腺管结构改变,间质水肿,炎细胞浸润,纤维组织增生,而BMSCs治疗组大鼠前列腺炎症明显减轻。PCR和ELISA分析显示ABP组IL-1βmRNA(0.829±0.121)、蛋白(271.75±90.59)pg/ml和TNF—αmRNA(0.913±0.094)、蛋白(105.78±19.05)pg/ml含量均显著高于对照组[(0.3424±0.087)、(45.76±17.99)pg/m1]、[(0.247±0.054)、(19.42±7.75)pg,/ml](P均〈0.01)及BMSCs治疗组[(0.433±0.072)、(51.34±22.13)pg/m1]、[(0.313±0.076)、(28.38±8.78)pg/m1](P〈0.01);CBP组IL-1βmRNA(0.975±0.114)、蛋白(265.31±71.34)pg/ml和TNF-理mRNA(0.886±0.084)、蛋白(107.45±26.11)pg/ml含量也均显著高于对照组及BMSCs治疗组[(0.396±0.064)、(56.37±21.22)pg/m1]、[(0.417±0.068)、(29.21±10.22)pg/m1](P均〈0.01)。结论:注入BMSCs能够减轻大肠埃希菌引起的前列腺炎症反应,其作用可能与减少炎细胞浸润,降低IL-1β、TNF-α水平有关。
Objective: To investigate the inhibitory effect of bone marrow mesenchymal stem cells (BMSCs) on E coil-induced prostatitis in rats. Methods : BMSCs were isolated, cultured and amplified by the attached choice method. Fifty SD rats were random- ized into five groups of equal number: normal control, acute bacterial prostatitis (ABP), chronic bacterial prostatitis (CBP), ABP + BMSCs, and CBP + BMSCs, and the animals in the latter four groups were injected with E. coli into both sides of the prostate under ul- trasound guidance for 1 - 14 days to induce ABP and for 4 - 12 weeks to induce CBP. The control rats were injected with the same amount of PBS. Two weeks after injection of BMSCs into the prostates, patbomorphological changes in the prostate were observed under the light microscope and the mRNA and protein levels of IL-113 and TNF-ct determined by RT-PCR and ELISA, respectively, followed by statistical analysis with SPSS 18.0. Results : Histopathological evaluation showed typical pathological inflammatory changes in the prostates of the rats in the ABP and CBP groups, including glandular structural changes, interstitial edema, inflammatory cell infiltra- tion, and fibrous hyperplasia, which were all remarkably relieved after treated with BMSCs. The mRNA and protein levels of IL-1 β( [ 0.829 ± 0. 121 ] and [ 271.75 ± 90.59 ] pg/ml) and TNF-α ( [ 0.913 ± 0. 094 ] and [ 105.78 ± 19.05 ] pg/ml) in the ABP and those of IL-1 [3 ( [ 0. 975 ± 0. 114 ] and [ 265.31 ± 71.34 ] pg/ml) and TNF-c~ ( [ 0. 886 + 0. 084 ] and [ 107.45 + 26.11 ] pg/ml) in the CBP groups were significantly higher than those in the control rats ( [ 0. 342 ± 0. 087 ] and [ 45.76 ± 17.99 ] pg/ml, P 〈 0.05 ) ; ( [ 0. 247 ± 0. 054 ] and ( [ 19.42 ± 7.75 ] pg/ml, P 〈 O. 01 ) as well as than those in the ABP + BMSCs ( [ 0. 433 ± 0. 072 ] and [51.34±22.13] pg/ml, P〈0.05); ([0.313±0.076] and [28.38 ~8.78] pg/ml, P〈0.01) and the CBP+BMSCs group ([0.396±0.064] and [56.37±21.22] pg/ml, P〈0. 05) ; ([0.417±0.0681 and [29.21 ±10.22] pg/ml, P〈0.01). Con- clusion : Injection of BMSCs can reduce E coli-induced prostatic inflammation reaction, which may be associated with its reduction of inflammatory cell infiltration and the expressions of IL-1β and TNF-α in the prostate tissue.
