摘要
为了寻找有效提取杏鲍菇(Pleurotus eryngii Quel)菌丝胞外酶的方法,为杏鲍菇等食用菌胞外酶的活性研究和生长规律提供试验依据,研究通过p H 4.8的柠檬酸缓冲液、去离子水2种处理方式提取杏鲍菇菌丝胞外酶,并通过测定8种胞外酶酶活来对这2种方式进行比较分析。结果表明,2种提取方式对杏鲍菇菌丝胞外酶的酶活有差异,其中,酸性纤维素酶和果胶酶经酸处理酶活是水处理的2倍多;木聚糖酶和木质素降解相关酶类经酸处理的酶活也高于水处理,但淀粉酶和蛋白酶经酸处理酶活低于水处理。
In order to find effective methods of extracting extracellular enzymes from Pleurotus eryngii mycelium and provide reliable experimental basis for studying activity of extracellular enzyme and regulating growth of Pleurotus eryngii or other edible fungi, two approaches including acid(citrate buffer at p H 4.8) and distilled water process were used to extract extracellular enzymes from Pleurotus eryngii mycelium. Activities of eight extracellular enzymes were determined to compare the two methods. The results showed that the two extraction methods had significant differences in activity of extracellular enzyme. The activity of Pectinase and cellulase extracted with acid was twice as that of water. The activities of xylanase, pectinase and lignin related enzymes after acid treatment were higher than that of water. The activities of amylase and protease in acid extraction was lower than that in water extraction.
