摘要
MADS-box基因家族是一类具有特异性序列的转录因子,并在植物体发育过程中起着重要作用。本研究以毛白杨形成层为材料,利用RACE技术克隆得到4个MADS-box基因,命名为Pt MADS1、Pt MADS2、Pt MADS3和Pt MADS5。Pt MADS1全长1 067 bp,开放阅读框(ORF)长度678 bp,编码含有225个氨基酸;Pt MADS2全长912 bp,ORF长度609 bp,编码含有202个氨基酸;Pt MADS3全长909 bp,ORF长度654 bp,编码含有217个氨基酸;Pt MADS5全长1 029 bp,ORF长度663 bp,编码220个氨基酸。系统进化树分析表明Pt MADS1与SVP同源性较高,属于STMADS11亚族;Pt MADS2与AGL12基因同源性较高,属于AGL12亚族;Pt MADS3和Pt MADS5分别与AGL14和SOC1同源性较高,同属于SOC1亚族。
MADS-box gene family is a class of transcriptional factors having specific sequences, which plays diverse roles in plant development. Four MADS-box genes were isolated from cambium materials of Populus tomentosa by using RACE PCR strategy, named as Pt MADS1, Pt MADS2, Pt MADS3 and Pt MADS5, respectively.Pt MADS1 has 1067 bp in length, including an ORF of 678 bp that encodes a 225 amino acid peptide; Pt MADS2 having 912 bp including an ORF of 609 bp encodes a 202 amino acid peptide. Pt MADS1 having 909 bp, including an ORF of 654 bp encodes a 217 amino acid peptide and Pt MADS1 having 1 029 bp, including an ORF of 663 bp encodes a 220 amino acid peptide. Alignment and homology comparison and phylogenetic tree analysis showed that Pt MADS1 shares high homology with SVP in Arabidopsis, which belongs to STMADS11 subfamily; Pt MADS2 shares high homology with AGL12 in Arabidopsis, which belongs to AGL12 subfamily; Pt MADS3 and Pt MADS5 share high homology with AGL14 and SOC1 in Arabidopsis, both belong to SOC1 subfamily.
