摘要
目的:比较薄层扫描(TLCS)法与高效液相色谱(HPLC)法测定二至丸中齐墩果酸含量的方法。方法:分别采用TLCS法和HPLC法。TLCS法参照2010年版《中国药典》(一部)的方法,波长分别为530、680 mm。HPLC法采用色谱柱为ODS,流动相为乙腈-水(80∶20,V/V),检测波长为220 nm,流速为1 ml/min,柱温为25℃。结果:HPLC法中,齐墩果酸的进样量在0.200 68-1.003 4μg范围内与峰面积呈良好线性关系;精密度、稳定性、重复性试验的RSD〈2%;平均加样回收率为100.7%,RSD为0.7%(n=6)。TLCS法测得二至丸中齐墩果酸的含量为8.78 mg/g,HPLC法测得其含量为9.82 mg/g,二者比较差异无统计学意义(P〉0.05)。结论:该研究所建立的HPLC方法操作快速、简便,结果稳定、可靠,更适用于二至丸中齐墩果酸的含量测定。
OBJECTIVE:To compare the content determination method for oleanic acid in Erzhi pill by TLCS and HPLC.METHODS:TLCS and HPLC were conducted. Chinese Pharmacopoeia(first part,2010 edition)was used for TLCS. In HPLC,the wavelengths were respectively 530 and 680 mm;the column was ODS with the mobile phase of acetonitrile-water(80 ∶ 20,V/V)at the flow rate of 1 ml/min;the detection wavelength was 220 nm and the temperature was 25 ℃. RESULTS:In HPLC,there was a good linear relationship between the volume and the peak area in the range of 0.200 68-1.003 4 μg;the RSDs of precision,stability and repeatability was less than 2% and the average recovery was 100.7%(RSD=0.7%,n=6). TLCS showed that the content of oleanic acid in Erzhi pill was 8.78 mg/g and HPLC showed that the content was 9.82 mg/g,with no significant difference(P〉0.05). CONCLUSIONS:The established HPLC is stable,reliable,rapid,easy and feasible. It is more suitable for the content determination of oleanolic acid in Erzhi pill.
