摘要
目的探讨妊娠期糖尿病(gestational diabetes mellitus,GDM)与孕妇血清、脐血和胎盘组织中核转录因子NF-κBp65的相关性。方法收集2011年11月至2013年3月天津市中心妇产科医院157名足月妊娠孕妇及正常未孕女性的临床资料,其中GDM孕妇38例(Ⅰ组)、正常孕妇56名(Ⅱ组)、正常未孕女性63名(Ⅲ组)。采用酶联免疫吸附测定3组研究对象血清、脐血中NF-κBp65的表达,采用免疫组化法检测孕妇胎盘组织中NF-κBp65的表达,并进行比较分析。结果Ⅰ组母体血清和脐血中NF-κBp65水平均高于Ⅱ组(均P<0.05);Ⅱ组、Ⅲ组母体血清中NF-κBp65水平比较差异无统计学意义(P>0.05);Ⅰ组胎盘组织中的NF-κBp65表达阳性率100.0%(38/38)高于Ⅱ组的12.5%(7/56)(P<0.05)。结论 NFKBp65可能在GDM的病理生理过程中发挥了一定作用。
Objective To explore the correlation between gestational diabetes mellitus( GDM) and the nuclear transcription factor NF- κBp65 in maternal blood,umbilical cord blood and placental tissue. Methods Data of 157 pregnant women in The Department of Gynecology and Obstetrics Tianjin Central Hospital from Nov 2011 to Mar 2013 were collected. 38 pregnant with GDM( group Ⅰ),56 women with normal pregnancy( group Ⅱ) and 63 women of childbearing age( group Ⅲ) were enrolled. ELISA techniques was employed to measure the expression of NF- κBp65 in the blood serum and umbilical cord blood,and the expression of NF- κBp65 in placental tissue was measured by immunohistochemical staining. The results were compared between 3 groups. Results NF- kappa Bp65 level in maternal blood serum and cord blood of group Ⅰ were higher than group II( P〈 0. 05). NF- kappa Bp65 level in maternal blood serum and cord blood between group Ⅱ and group Ⅲ had no significant difference( P〈0. 05). The positive expression rate of NF- kappa Bp65 in placental tissue of group Ⅰ[100. 0%( 38 /38) ] was higher than group Ⅱ[12. 5%( 7 /56) ]( P〈0. 05). Conclusion NF- KBp65 may play a certain role in the pathophysiology of gestational diabetes mellitus.
