探究增生性瘢痕成纤维细胞中CDK4、CDK2、CDK1基因与细胞周期的相关性 被引量:2

摘要 目的:探究与分析增生性瘢痕成纤维细胞中CDK4、CDK2、CDK1基因及蛋白的表达与细胞周期之间的相关性。方法:选取本院自2012年7月-2014年7月采集的40份增生性瘢痕标本,按照增生性瘢痕的时间段将所采集的标本进行分组,共分为3个月组、6个月组、1年组及2年组,每组各10份标本,另选择同时期采集到的10份正常标本作为常规对照组。观察与对比各个标本中CDK4、CDK2、CDK1基因及蛋白情况,同时对成纤维细胞周期进行检测。结果:3个月组、6个月组标本中CDK4、CDK2、CDK1基因含量及蛋白含量与对照组相比差异均有统计学意义(P<0.05);6个月、1年组、2年组标本中CDK4、CDK2、CDK1基因含量及蛋白含量与3个月组相比差异均有统计学意义(P<0.05);3个月、1年组、2年组标本中CDK4、CDK2、CDK1基因含量及蛋白含量与6个月组相比差异均有统计学意义(P<0.05)。增生期瘢痕早期高比例在细胞分裂周期中的S期间及G2/M期,结果可见,针对此细胞周期进行相关的基因调控干预,可对增生性瘢痕进一步发生发展起到有效抑制作用。结论:处于不同增生性瘢痕的时期中CDK4、CDK2、CDK1基因及蛋白的表达趋势大致相符,增生性瘢痕细胞周期的分布情况与上述三个蛋白执行功能的情况呈相互对应的关系,为临床研究提供可靠依据。 Objective:To explore and analyze CDK4,CDK2 and CDK1 gene in the hyperplastic scar fibroblasts,protein expression and the relationship with the cell cycle.Method:40 of hyperplastic scar samples in our hospital were selected from July 2012 to July 2014,according to the period of hyperplastic scar were divided into the 3 months group, the months group,the 1 year group and the 2 years group,each group with 10 specimens.10 normal specimens were collected at the same time as a normal control group.The samples of CDK4,CDK2,CDK1 gene and their protein were observed and compared,at the same time the fibroblasts cycle were tested. Result:The content of CDK4,CDK2,CDK1 gene and protein in the 3 months group,and 6 months group compared with the control group,the differences were statistically significant(P〈0.05);the content of CDK4,CDK2,CDK1 gene and protein in the 6 months group,1 year group,2 years group compared with 3 months group,the differences were statistically significant(P〈0.05);the content of CDK4,CDK2,CDK1 gene and protein in the 3 months group,1 year group,2 years group compared with 6 months group,the differences were statistically significant(P〈0.05).Early phase hyperplasia scar accounted for high percentage during cell division cycle in S and G2/M phase,the result showed regulatory intervention with the related gene for the cell cycle can further develop hyperplastic scar effective inhibition effect. Conclusion:In different periods the trend of gene of CDK4,CDK2,CDK1 and protein expression is broadly in line.The distribution of cell cycle corresponds the above three protein executive function each other,which provide reliable basis for clinical study.
