摘要
目的研究广金钱草提取物对α-异硫氰酸萘酯(ANIT)诱导急性肝内胆汁淤积大鼠的保护作用。方法SD大鼠共分7组:正常对照组,模型组,阳性对照(熊去氧胆酸)组,广金钱草水煎液组,广金钱草正丁醇萃取物高、中、低剂量组。建立ANIT胆汁淤积大鼠模型,除正常对照组和模型组外,其余各组分别灌胃给予相应药物干预。大鼠手术插管收集胆汁后,腹主动脉取血。测定给药后各时间点胆汁流量,并检测血清中谷丙转氨酶(GPT)、谷草转氨酶(GOT)、碱性磷酸酶(ALP)、总胆红素(TB)、直接胆红素(DB)、总胆汁酸(TBA)、γ-谷氨酰转肽酶(γ-GT)、一氧化氮(NO)、环磷酸腺苷(cAMP)、谷胱苷肽(GSH)水平及胆汁中TBA、GSH水平。结果与模型组比较,广金钱草正丁醇萃取物高、中、低剂量能显著增加胆汁淤积大鼠的胆汁分泌量(P<0.01),显著降低血清中GPT、GOT、ALP、TBA水平,同时升高NO、cAMP、GSH水平(P<0.01,P<0.05);并能显著增加胆汁中TBA水平(P<0.01),降低胆汁中GSH水平(P<0.01)。结论广金钱草具有抗ANIT诱导的急性胆汁淤积、保护肝脏及促进胆汁分泌的作用。
Objective To study the protective effect of Herba Desmodii Styracifolii(HDS) on rat acute cholestasis induced by alpha- naphthylisothiocyanate(ANIT).Methods SD rats were randomized into normal control group,model group,ursodeoxycholic acid group,HDS water- extract group,and high-,middle- and low- dose HDS n- butanol extract groups.The rat cholestasis model was established and then was intervened by ursodeoxycholic acid,HDS water- extract,HDS n- butanol extract,respectively.After collecting the bile by the operation,the rats were executed for blood sampling.Bile flow of the rats was detected at different time points after medication,and we also measured serum levels of glutamic pyruvic transaminase(GPT),glutamic oxalacetic transaminase(GOT),alkaline phosphatase(ALP),total bilirubin(TBIL),total bile acids(TBA),gamma glutamyl transpeptidase(γ- GT),nitric oxide(NO),cyclic adenosine monophosphate(c AMP),and glutathione(GSH) as well as TBA and GSH levels in the bile.Results Compared with the model group,high-,middle-,and low- dose HDS n- butyl alcohol showed an obvious effect on increasing the bile flow(P〈0.01),decreasing serum GPT,GOT,ALP and TBA levels,increasing serum NO,c AMP,and GSH levels(P〈0.01,P〈0.05),enhancing bile TBA level(P〈0.01)and reducing bile GSH level(P〈0.01).Conclusion HDS exerts an effect on counteracting ANIT- induced acute cholestasis in the rats,protecting liver cells and promoting the bile flow.
