摘要
【目的】研究不同来源的不育系是否具有相似的遗传背景和相同的不育候选基因。【方法】利用PCR,RT-PCR技术,以及Northern blot技术分析orf160及其周边序列在104-7A,湘远A,晋A,HAMS277等棉花不育衍生系线粒体中存在状况及表达。【结果】发现orf160在被调查的CMS系中均存在,且均有转录,而且在P30A(104-7A的衍生系)中与其上游atp4基因间均为单独转录;同时发现orf160在被调查的CMS系中具有相似的旁侧序列。【结论】被调查的CMS系线粒体均含有orf160基因及基本一致的旁侧序列;作为不育候选基因的orf160,与其上游的atp4基因不存在共转录现象。
【Objective】The purpose of project aims to research whether the different sources in sterile lines have similar genetic background and the same candidate gene for infertility. 【Method】In this paper,PCR,RT-PCR technology,and Northern blot analysis of the technology and its surrounding orf160 sequence of infertility in 104-7A,far Xiang A,Jin A,HAMS277 mitochondrial derivatives such as cotton and conditions that exist in the expression were used. 【Result】It was found that orf160 existed in CMS lines that had been tested,and were transcribed,and its rooms were individually upstream atp4 gene transcription in P30A( derived lines of 104-7A); orf160 CMS was also found to have similar flanking sequences in the surveyed lines.【Conclusion】CMS mitochondrial orf160 surveyed contained basically the same gene and flanking sequences as infertility candidate gene orf160,and its atp4 gene upstream of the transcription phenomenon did not exist.
