校正超滤法测定木香烃内酯和去氢木香内酯的大鼠血浆蛋白结合率 被引量:3

摘要 目的:同时测定木香烃内酯和去氢木香内酯的大鼠血浆蛋白结合率.方法:采用加标血浆样品测定超滤管对药物的非特异吸附系数,对超滤法进行校正,并用已知血浆蛋白结合率的药物紫杉醇验证、校正超滤法的准确性.结果:测得木香烃内酯大鼠血浆蛋白结合率为88.25% ~ 92.14%,去氢木香内酯大鼠血浆蛋白结合率为85.34%~89.37%.结论:木香烃内酯的血浆蛋白结合率略高于去氢木香内酯,该校正超滤法可以广泛应用于药物的血浆蛋白结合率测定. Objective: To determinate costunolide and dehydrocostus lactone plasma protein binding rate in rat plasma. Methods: The spiked plasma was used to determine nonspecific binding coefficient of drugs with the ultrafiltration device, and then the nonspecific binding coefficients were used for rectifying the determination results of plasma protein binding rates of drugs. The accuracy of this method was validated by known plasma protein binding rates of paclitaxel. Result: The rat plasma protein binding rate of costunolide and dehydrocostus lactone were 88.25%-92. 14% and 85.34%-89.37%, respectively. Conclusion: The rat plasma protein binding rate of costunolide is slightly higher than that of dehydrocostus lactone, and this rectified ultrafiltration can be widely used for determining plasma protein binding rate of drugs.
