摘要
目的:在通过观察紫杉醇对体外培养大鼠肝细胞的影响来探讨紫杉醇自身毒副作用.方法:采用四唑盐(MTT)比色法及形态学方法观察紫杉醇在不同质量浓度下作用不同时间对肝细胞的影响;采用Hoechst 33258荧光染色法观察紫杉醇作用后,肝细胞的形态变化.结果:质量浓度分别为5、10、20、40、80μg/L的紫杉醇分别作用24、48、72 h后,随着质量浓度的增加、作用时间的延长对体外培养的肝细胞损伤加大、抑制率增加;相同质量浓度的紫杉醇不同作用时间的抑制率无时间效应关系;24 h的IC50质量浓度为28.5μg/L,并能诱导肝细胞发生凋亡.结论:紫杉醇在一定的质量浓度和长时间作用下对肝细胞有细胞毒作用.
Aim:Observing the side-effects and toxicity of taxol on rat hepatocytes in order to provide with experimental data for clinical reference.Methods:The effects of taxol at different concentrations and with different treating time on hepatocytes were observed by using MTT assay and morphology.Ho-echst 33258 fluorescence method was employed to analyze the morphological changes in hepatocytes. Results:After the hepatocytes were treated with taxol at different concentrations(5、10、20、40、80 μg/L) for 24 h、48 h、72 h,the cells were found to be damaged and growth inhibited increasingly with the con-centration of taxol and the time of treatment.However,there was no time-dependent manner at a same dose of taxol.The IC50 of taxol against hepatocytes was found to be 28.5 μg/L,and taxol also induced ap-optosis of hepatocytes.Conclusion:Taxol has toxicity on hepatocytes in a range of concentrations and af-ter a certain time of action.
