冠心宁注射液中酚酸类成分的质量分析 被引量:7

Simultaneous determination of phenolic acids in Guanxinning injection by HPLC

摘要 通过高效液相色谱法同时测定冠心宁注射液中丹参素、原儿茶醛、迷迭香酸、紫草酸、丹酚酸B的含量。采用Kroma-si C_18色谱柱(4.6mm×250mm,5μm),以乙腈-3%甲酸水溶液为流动相,流速1mL·min^-1,柱温30℃,检测波长280nm。结果显示,丹参素、原儿茶醛、迷迭香酸、紫草酸、丹酚酸B分别在进样量0.00600~4.00,0.00615~4.10,0.00600~4.00,0.00606~4.04,0.00609~4.06μg与峰面积呈良好的线性关系,r分别为0.9993,0.9994,0.9993,0.9991,0.9992;回收率(n=6)分另0为98.9%(RSD0.75%),98.1%(RSD1.2%),100%(RSD0.77%),98.7%(RSD1.7%),102%(RSD0.68%)。按所建方法对13批样品中上述5种成分的含量进行测定。该方法简便、准确,重复性好,可用于冠心宁注射液中丹参素、原儿茶醛、迷迭香酸、紫草酸、丹酚酸B的质量控制。 This paper was aim to determine five phenolic acids, sodium danshensu (SD), protocatechnic aldehyde (PA), rosma- rinic acid (RA), lithospermic acid (LA) and salvianolic acid B (SAB), in Guanxinning injection. In the test, Kromasil C18 column (4. 6 mm× 250 mm,5 μm) was adopted, with acetonitrile-3 % formic acid solution as the mobile phase for gradient elution. The flow rate was 1 mL· min^- 1 , the column temperature was 30 ℃ and the detection wavelength was 280 nm. According to the results of the test, SD, PA, RA, LA and SAB showed good linear relations between peak areas and sample sizes in 0. 006 06-4. 04(r =0. 999 3) , 0. 006 15-4. 10 ( r = 0. 999 4) ,0. 005 94-3.96 ( r = 0. 999 3 ), 0. 006 06-4.04 ( r = 0. 999 1 ) and 0. 006 09-4. 06 ( r = 0. 999 2 ) p,g, re- spectively. The average recoveries (n = 6) were 98.9% (RSD 0. 75 % ), 98. 1% (RSD 1.2% ), 100% (RSD 0. 77 % ) , 98. 7 % ( RSD 1.7% ) ,102% (RSD 0. 68% ), respectively. The above 5 components were determined in 13 hatches of samples by using the estab- lished method. The method was simple, accurate and highly reproducible that it could be used for quality control of the components in Guanxinning injection.
