高迁移率族蛋白B1活化p38 MAPK对人支气管上皮细胞胸腺基质淋巴生成素表达的影响 被引量:1

摘要 目的探讨高迁移率族蛋白B1(HMGB1)对人支气管上皮细胞(HBE)胸腺基质淋巴生成素(TSLP)表达的影响及机制。方法应用实时荧光定量PCR和ELISA分别检测HMGB1刺激HBE后TSLP基因表达及蛋白释放水平;用Western blot和ELISA分别检测p38 MAPK抑制剂SB202190干预HBE后磷酸化ATF-2蛋白(pATF-2)表达及TSLP释放水平。结果 (1)HMGB1以浓度依赖形式增加HBE TSLP的表达;(2)相对于HMGB12 000 ng/m L的干预,SB202190抑制剂可明显减少活化的HBE p-ATF-2蛋白的表达(P<0.01)及其TSLP的释放(P<0.01)。结论 HMGB1可活化p38 MAPK通路增加HBE TSLP的表达,可能以此参与哮喘的发病过程。
