摘要
目的:研究复方丹参片对大脑中动脉缺血再灌注所致血管性痴呆大鼠的影响。方法:复制大鼠中动脉缺血再灌注模型,采用避暗实验及水迷宫实验观察复方丹参片对学习、记忆能力的影响,测定SOD活性、MDA含量、及Ach E含量,观察复方丹参片对脑组织损伤的影响。结果:复方丹参片组(0.225g/kg、0.45g/kg、0.9g/kg)VD大鼠的学习记忆能力显著改善。复方丹参片0.9g/kg脑组织的SOD活性升高,MDA含量降低;复方丹参片0.45g/kg MDA含量降低;复方丹参片0.225g/kg SOD活性升高,氧化应激损伤显著减轻。复方丹参片0.225g/kg VD大鼠脑组织的CA1-CA3海马区神经细胞数目减少的情况有所改善;复方丹参片0.45g/kg、0.9g/kg大鼠脑组织神经细胞数目减少、变性坏死、软化灶形成均有显著的改善。结论:复方丹参片对大脑中动脉缺血再灌注所致血管性痴呆大鼠脑损伤及学习记忆功能具有明显的改善作用。
Objective: To observe the effect of Compound Danshen Tablets( CDT) on rat with vascular dementia caused by ischemia and reperfusion in middle cerebral artery. Method: 75 rats were randomly divided into 6 groups,including control group,model group,sham group,CDT Ⅰ group( 0. 225 g / kg),CDT Ⅱ group( 0. 45 g / kg),CDT Ⅲ group( 0. 9g / kg),and co-dergocrine mesylate group( 0. 00625 g / kg).The vascular dementia rat model was established by Zealonga method. The effects on learning and memory abilities of rats were observed by channel water maze task and step-through task. The SOD activity,contents of MDA and Ach E were determined to explore the effects on brain injury. Results: Abilities of learning and memory were improved significantly in CDT groups( 0. 225 g / kg,0. 45 g / kg,0. 9g / kg). Injuries caused by oxidative stress were induced objectively: activity of SOD was increased and concentration of MDA was decreased in CDT Ⅲ group( 0. 9g/kg),concentration of MDA was decreased in CDT Ⅱ group( 0. 45g/kg),and activity of SOD was increased in CDT Ⅰ group( 0.225 g / kg). Degeneration and necrosis of nerve cells in liippocampus were improved significantly in CDT groups( 0. 45 g / kg,0. 9g / kg). Conclusion: The brain injuries caused by ischemia and reperfusion in middle cerebral artery of vascular dementia rats could be improved by CDT.
