摘要
目的了解江苏部分地区动物中发热伴血小板减少综合征病毒(SFTSV)携带状况,探讨其可能的传播媒介和自然宿主。方法 2012年3月-12月在江苏省发现发热伴血小板减少综合征病例的7个市/县,采集鼠、牛、羊、刺猬、鸟类和蜱等动物标本,提取总核酸后,应用实时荧光RT-PCR法对标本进行布尼亚病毒S基因核酸的检测,阳性标本采用Vero细胞进行病毒分离。结果采集761只(管)动物,43只(管)检出SFTSV核酸阳性,阳性率为5.65%。采集3 728份标本,阳性49份,阳性率为1.31%。鼠的病毒携带率最高(6.84%),蜱标本阳性率最高(4.94%)。除蜱标本外的3 647份标本中检出阳性45份,阳性率为1.23%,不同器官阳性率分别为淋巴结(50.00%)、血清(2.34%)、脾(1.64%)、心(1.64%)、肝(1.28%)、肺(1.09%)、脑(0.94%)、肾(0.73%)、血块(0.41%)。5月标本阳性率最高(4.01%)。从1份羊血清及其体表1份蜱标本中各分离出1株SFTSV病毒。结论鼠、牛、羊和蜱均可携带SFTSV,可能是其自然宿主或传播媒介。
Objective To investigate the severe fever with thrombocytopenia syndrome virus(SFTSV)-carrying status of animals in some regions of Jiangsu province,and to study its possible media and natural hosts.Methods Specimens from mice,cattle,goats,hedgehogs,birds and ticks were collected from 7cities/counties of Jiangsu province,where SFTSV patients were identified between Mar.to Dec.2012.Total nucleic acid was extracted from the above specimens and subjected to real-time RTPCR for detection of SFTSV S gene.Vero cells were used for virus isolation for positive samples.Results Among 761 animals,43were positive for the nucleic acid of SFTSV,resulting in a positive rate of 5.65%.A total of 3 728 specimens were collected,among which 49 were positive,resulting in a positive rate of 1.31%.Mice had the highest virus-carrying rate among all animals(6.84%),while tick specimens had the highest virus-carrying rate among all specimens(4.94%).Forty-five of 3 647 specimens from which tick specimens were excluded from were found to have the nucleic acid of SFTSV,resulting in a positive rate of1.23%.Positive rates of different organs in descending order were lymph node(50.00%),serum(2.34%),spleen(1.64%),heart(1.64%),liver(1.28%),lung(1.09%),brain(0.94%),kidney(0.73%),and blood clots(0.41%).The positive rate of specimens was highest in May(4.01%).Two SFTSV virus strains were isolated from one goat serum sample and one tick stuck on the skin of above goat.Conclusion Mice,cattle,goats and ticks can carry SFTSV,and they may be the natural hosts or media of SFTSV.
