摘要
目的:建立适宜于抗抑郁药物评价的抑郁大鼠动物模型及综合评价体系,为抗抑郁药物的药理药效评价奠定前期基础。方法:选用SD雄性大鼠,在CUMS法基础上,建立抑郁大鼠动物模型,考察造模前和造模7天、14天、21天、35天时大鼠体重、悬尾次数、糖水消耗、血清皮质酮等指标的变化,综合评价优选造模条件。结果:与空白组比较,造模21天模型组在体重、糖水消耗、悬尾次数、游泳时间、敞箱实验等方面均有明显降低趋势,大鼠血清皮质酮含量明显减少,实验结果证明了抗抑郁药物评价的抑郁大鼠动物模型建立的合理性。结论:该模型克服以往存在的工作量大,持续时间长、指标评价单一等问题,为抗抑郁药物的药效评价奠定实验基础。
Objective: To establish a depression animal model of rats that is suitable for antidepressant drug evaluation and a com- prehensive evaluation system, the model laid the foundation for evaluation of the pharmacological efficacy of antidepressant drugs. Methods:On the basis of CUMS method, to establish a depression animal model in SD male rats, and Before the investigation mod- eling, modeling 7 days, 14 days, 21 days, 35 days rat body weight, hanging tail number, sugar consumption, and other indicators of changes in serum corticosterone, comprehensive evaluation of the preferred modeling conditions. Results: Compared with the control group, model group in body weight, sugar consumption, hanging tail number, swimming time, Open Field, etc. are significant changes, and significantly reduced in cortieosterone levels in the blood of rats . The experimental results prove thai the depression animal model used of to evaluate antidepressant drug is rationally. Conclusion: This model overcomes the heavy workload, long duration, a single index evaluation and other issues in the past, the study evaluated the efficacy of antidepressants experimental basis.
