摘要
目的 研究胎盘多肽对骨髓病态造血的影响.方法 选取2010年10月~2012年12月住院治疗的30例骨髓增生异常综合征(MDS)患者作为研究对象,另选取30例同期体检的健康人作为对照组.采用免疫组化方法检测其造血细胞[骨髓细胞(Bone marrow cell Fas ligand,Fas)和骨髓细胞配体(Bone marrow cell Fas ligand,FasL)]的表达.体外培养使用胎盘多肽,并观察其对两组造血功能以及造血细胞凋亡的影响.结果 MDS患者不同病种的Fas表达不同,幼红细胞性顽固性贫血(refractory anemia,erythroblastic,RAEB)患者明显低于难治性贫血(Refractory anemia,RA)患者,过量的幼红细胞性顽固性贫血(refractory anemia with excess blasts,RAEB-t)患者明显低于RAEB患者,三者之间差异均具有统计学意义(均P<0.05).MDS患者的CFU-GM和CFU-E明显低于对照组(P<0.05),加入胎盘多肽对对照组的粒细胞-巨噬细胞集落单位(Granulocyte macrophage colony Generating units,CFU-GM)和红细胞集落生成单位(Red cellsColony forming unit,CFU-E)无明显影响,但明显升高MDS患者的CFU-GM和CFU-E(P<0.05),浓度越大,升高越明显.MDS患者造血细胞凋亡明显高于对照组,加入胎盘多肽后明显降低MDS患者的细胞凋亡.结论 MDS患者的Fas和FasL的表达异常,胎盘多肽可能通过降低造血细胞凋亡从而促进正常造血.
Objective To study the influences of placenta polypeptide to marrow hemopoiesis. Methods 30 pa- tients with myelodysplastic syndrome (MDS) were selected as the research objects, and another 30 healthy people who had physical examinations in our hospital in the corresponding period were selected as the control group. Immunohisto chemical method was used to detect and compare the expression of Chinese name (Bone marrow cell Fas ligand Fas Fas) and (Bone marrow cell Fas ligand FasL) expression, in hematopoietic cells. Placenta polypeptide was cultured in vitro and its influences to hematopoietic function and hematopoietic cell apoptosis for the two groups needed to be observed. Results The expression of Fas in different diseases entities for MDS patients was different; the expression for RAEB pa- tients was significantly lower than that of Chinese name (Refractory anemia,RA) patients, while the expression for Chi- nese name (refractory anemia with excess blasts RAEB-t ) patients was obvious lower than that of RAEB patients; the differences among these were of statistical significance (P〈0.05). For MDS patients, their Chinese names (Granulocyte macrophage colony Generating units CFU-GM) and (Red cellsColony forming unit CFU-E)were significantly lower than that of the control group, and there existed no obvious influence to CFU-GM and CFU-E for people in the control group when adding placenta polypeptide, while the CFU-GM and CFU-E for MDS patients were obviously higher, and if the concentration was greater, the higher would be more obvious. The apoptosis of hematopoietic cell for MDS patients was significantly higher than that of the control group, and adding the placenta polypeptide had reduced the apoptosis signifi- cantly for MDS patients. Conclusion The expressions of Fas and FasL for MDS patients were abnormal; and the plaeen ta polypeptide could strength the hemopoiesis of MDS patients through reduce the aoootosis.
