菊苣对高尿酸血症鹌鹑肾脏有机阴离子转运体OAT3-LIKE的影响 被引量:8

Effect of Chinese Herb Chicory on Renal Organic Anion Transporter OAT3-LIKE of Hyperuricemia Quails

摘要 目的从肾脏转运蛋白角度探究中药菊苣降尿酸的药效机制。方法将40只雄性迪法克鹌鹑按体质量随机分为5组,正常组给予普通饲料,其余各组均给予含酵母饲料(15 g·kg-1·d-1)建立高尿酸血症模型,同时给药组分别给予菊苣5,7.5 g·kg-1·d-1,苯溴马隆20 mg·kg-1·d-1,实验第35天处死。检测鹌鹑血清尿酸(Uric Acid,UA)、尿素氮(Urea Nitrogen,BUN)水平,并用Real-time RT-PCR(Real-time Reverse TranscriptionPolymerase Chain Reaction)方法检测其肾脏有机阴离子转运体OAT3-LIKE(Organic Anion Transporter3-LIKE)的mRNA表达水平,同时测定肝脏尿酸代谢相关酶活性。结果与正常组比较,实验第21天、28天、35天模型组鹌鹑血尿酸升高,第35天肾脏OAT3-LIKE m RNA表达水平下降。与模型组比较,实验第35天,菊苣可以降低鹌鹑血尿酸水平,上调其肾脏OAT3-LIKE mRNA表达水平并降低肝脏腺苷脱氨酶(ADA)活性。结论高嘌呤食饵可以成功诱导鹌鹑高尿酸血症;菊苣具有抑制高尿酸血症鹌鹑尿酸生成和促进尿酸排泄的双重作用;菊苣药效机制可能与降低ADA活性及上调OAT3-LIKE表达水平相关。 Objective To explore the pharmacodynamic mechanism of chicory on lowering uric acid from the aspects of renal transport protein. Methods Forty male quails were randomly divided into five groups. The normal group was fed with common forage, and the other groups were fed with forage containing yeast(15 g·kg^-1·d^-1) to establish hyperuricemia model. At the same time,the medication groups were given chicory in the dosage of 5,7.5 g·kg^-1·d^-1,and benzbromarone 20 mg·kg^-1·d^-1,respectively. The quails were sacrificed on the 35 thday. We detected serum uric acid(UA) and urea nitrogen levels and hepatic adenosine deaminase(ADA) activity,and examined the expression levels of renal organic anion transporter protein OAT3-LIKE mRNA using real-time quantitative reverse transcription-polymerase chain reaction(RT-PCR)method. Results On the 21 st,28th,35 thexperimental day,the UA level of the model group raised significantly than the normal group. On the 35 thday,OAT3-LIKE m RNA expression level of the model group was lower than that of the normal group,and chicory showed an effect on decreasing serum UA level,up-regulating renal OAT3-LIKE mRNA level and reducing renal ADA activity. Conclusion High purine diet can induce hyperuricemia in quails, and chicory has an effect on inhibiting UA formation and promoting UA secretion in hyperuricemia quails, which is probably related with the decrease of renal ADA activity and with the up-regulation of renal OAT3-LIKE mRNA expression.
