摘要 [目的]对北方黄酒麦曲中真菌的5.8S r DNA-ITS序列进行PCR扩增、测序以鉴定其中的真菌。[方法]利用梯度稀释法和划线纯化法共分离纯化得到5株霉菌和1株酵母,并对分离得到的真菌进行基因组DNA提取获得模板,用特异性引物进行PCR扩增,得到真菌的5.8Sr DNA-ITS片段,片段测序后进行Blast比对鉴定,构建系统发育树。[结果]将5株霉菌鉴定为多枝横梗霉(Lichtheimia ramosa),米曲霉(Aspergillus oryzae),产紫青霉(Penicillium purpurogenum),杂色曲霉(Aspergillus versicolor),交链孢霉(Alternaria mali),将一株酵母鉴定为酿酒酵母(Saccharomyces cerevisiae)。[结论]利用分子生物学的方法对北方黄酒麦曲样品中的真菌进行了分离、纯化和鉴定。 [ Objective ] 5.8S rDNA-ITS fragments of fungi in the wheat starter from north yellow millet wine were amplified and sequenced in order to identify the fungi. [ Method ] Five strains of moulds and one yeast strain were separated and purified by gradient dilution method and crossed purification. The genomic DNA of fungi cells were extracted as the PCR template, and 5.8SrDNA-ITS fragments of fungi were ampli- fied and sequenced respectively for blasting identify and phylogenetic tree construction. [ Result ] The moulds were identified as Lichtheimia ramosa, AspergiUus oryzae, PeniciUium purpurogenum, Aspergillus versicolor, Alternaria mali,and one yeast strain was identified as Saccharo- myces cerevisiae. [ Conclusion] Fungi in the wheat starter from north yellow millet wine were separated, purificated and identificated.
