摘要 [目的]研究影响胚乳愈伤组织诱导的因素,为三倍体植株再生奠定基础。[方法]以黑果腺肋花楸种子成熟胚乳为试材,研究取材时期、培养基种类、激素种类等因素对愈伤组织诱导的影响。[结果]取转色期的胚乳在MS+TDZ 0.05 mg/L+IBA 2.0 mg/L+蔗糖40 g/L+琼脂8 g/L的培养基上愈伤组织长势最好,诱导率最高可达92%,颜色为黄绿色且结构致密。[结论]适宜黑果腺肋花楸胚乳愈伤组织诱导的培养方法为转色期取材,光照强度为800 lx,培养基配方为MS+TDZ 0.05 mg/L+IBA 2.0 mg/L+蔗糖40 g/L+琼脂8g/L。 [ Objective ] The aim was to study factors affecting the endosperm callus induction,in order to lay a foundation for triploid plant regen- eration. [ Method ] The seeds of Arortia melanocarpa Elliot were used as experimental materials on this study to explore the influences of sampling time,different medium and hormone's concentrations on endosperm callus induction. [ Result ] The experiments showed that endosperm inoculated on MS medium supplemented with 0.05 mg/L TDZ,2.0 mg/L IBA gave highest callus induction rate 92% . The callus was yellow green with a compact structure. [ Conclusion ] The suitable medium for the induction of endosperm callus of Aronia melanocarpa Elliot was MS + TDZ 0.05 mg/L + IBA 2.0 mg/L +40 g/L + sucrose AGAR 8 g/L with light intensity of 800 lx in turning green period .
