益气活血药对舒张功能不全大鼠心功能及钙转运的干预研究 被引量:4

摘要 目的:研究益气活血药对舒张功能不全大鼠心肌细胞心功能及钙转运的干预作用。方法:建立压力负荷致舒张功能不全大鼠模型,按干预因素分为假手术组、模型组、益气组、活血组、益气活血组。造模后给药8周检测大鼠心功能,分离心肌细胞,检测心肌细胞收缩、舒张功能及钙瞬变、胞内钙浓度。结果:益气活血组舒张早/晚期峰值血流速度(E/A)较模型组升高;各给药组舒张50%时间(TR50)较模型组均显著降低,其中益气活血组效果最明显;活血组、益气活血组钙瞬变衰减常数(Tau)较模型组缩短;益气活血组舒张期钙浓度较模型组降低。结论:益气活血药通过改善心肌细胞钙转运能力,提高心肌细胞舒张功能,改善心脏舒张功能,并降低心衰心肌细胞钙浓度,从而预防心衰室性心律失常的发生。 Object: To explore the effects of reinforcing qi and promoting blood circulation on cardiac function and calcium transport in rats with diastolic dysfunction. Methods: The rat model of diastolic dysfunction was established by pressure-overload. According to the intervention, the rats were randomly divided into 5 groups of sham-operation, control, reinforcing qi ( RQ), promoting blood circulation ( PBC ), reinforcing qi and promoting blood circulation (RQ&PBC). Cardiac function was detected after 8 weeks of intervention, then cardiomyoeytes were isolated. The cardiomyocyte contractility and calcium transport was tested. Results: E/A ratio was higher in RQ&PBC group than in control group; Compared with control group, 50% time of relaxtion was reduced in RQ group, PBC group and RQ&PBC group, among which the effect of RQ&PBC group was the most significant; Tau were lower in PBC group and RQ&PBC group than in control group; Compared with control group, the diastolic calcium concentration of RQ&PBC group was reduced. Conclusion: The treatment of reinforcing qi and promoting blood circulation improved the ability of calcium transport and relaxtion in cardiomyocytes, leading to increased cardiac diastolic function and reduced cardiomyocyte calcium concentration, thus protected heart against ventricu]ar arrhythmia.
