摘要
目的研究紫外线对实验室污染的基因物质清除效果。方法采用基因扩增检测技术,对紫外线灯照射法清除基因物质污染的效果进行实验室评价。结果紫外线在辐射强度为60μW/cm2条件下照射6 h,对基因物质基本没有清除效果。将紫外线灯照射距离缩短到30 cm或延迟照射时间至12 h,亦未显示出对核酸污染物明显的清除效果。结论用紫外线清除实验室中的基因物质污染效果不明显。
Objective To study the clearance effect on DNA contamination sourced from genetically modified organism (GMO) detection laboratory by ultraviolet light irradiation. Methods Polymerase chain reaction was used to evaluate the clearance effect on exogenous gene contamination from GMO using ultraviolet lamp irradiation. Results There was no ob- vious scavenging effect on nucleic acid contaminants irradiated continually for 6 h under the condition of 60 μW/cm2 radia- tion intensity. While the irradiation distances of ultraviolet lamp was shortened to 30 cm or the irradiation time was delayed to 12 h, we did not find obvious removal effect of nucleic acid pollutants. Conclusion The removal effect of nucleic acid contamination by use of ultraviolet lamp irradiation in GMO detection lab is not obvious.
