摘要 目的建立HPLC法同时测定荷叶中金丝桃苷、异槲皮苷和槲皮素。方法荷叶甲醇提取液采用Agilent Eclipse XDB C18柱(250 mm×4.6 mm,5μm)分析;流动相为磷酸盐缓冲液(p H=4.0)-乙腈,梯度洗脱;体积流量1.0m L/min;检测波长356 nm;柱温30℃。结果金丝桃苷、异槲皮苷和槲皮素分别在0.041 6~0.416 0μg、0.038 0~0.380 0μg和0.008 8~0.088 0μg范围内线性关系良好,平均回收率分别为98.7%、98.5%和100.4%,RSD分别为1.1%、1.4%和2.2%。结论该方法操作简便准确,重复性较好,可为荷叶中黄酮类化合物的开发利用提供质量评价依据。 AIM To establish an HPLC method for simultaneous determining hyperoside, isoquercitrin and quercetin from Nelumbinis Folium. METHODS The separation of N. Folium methanolic extract was achieved on an Agilent Eclipse XDB C18 column (250 mm × 4. 6 mm, 5 μm). The mobile phase consisted of acetonitrile- phosphate buffer solution ( pH = 4.0 ) in a gradient elution mode at the flow rate of 1.0 mL/min. The detection wavelength was set at 356 nm, and the column temperature was maintained at 30 ℃. RESULTS Good linearities of hyperoside, isoquereitrin and quercetin were obtained within the ranges of 0. 041 6 - 0. 416 0, 0. 038 0 - 0. 380 0, and 0. 008 8 -0. 088 0 p^g, respectively. Their average recoveries were 98.7% , 98.5% and 100. 4% , and the RSDs were 1.1% , 1.4% and 2. 2% , respectively. CONCLUSION The method is simple, accurate and reproducible, which can provide a basis for exploitation and utilization of N. folium.
