摘要
采用双波长法对杂交谷及其双亲直链淀粉和支链淀粉的含量进行测定,并将父本和杂交谷子粒淀粉含量进行了比较,以评价父本对杂交谷淀粉含量的影响。结果表明,通过回归方程计算得到供试样品的直链淀粉与支链淀粉的含量分别为11%~15%和45%~65%;精密度试验和加标试验结果显示,每一样品5次重复测定结果的RSD〈1%,样品回收率为95.46%-102.23%,表明该方法稳定性好、重复性高,可用于小米淀粉含量的测定。通过相关性分析可知父本对杂交谷子粒淀粉含量的遗传效应。
The dual-wave length spectrophotometry method was used to determine starch content in hybrid millet and its parents, the effect of male parent on starch content of hybrid millet was evaluated by comparing the starch content of hybrid millet to male parent. The results showed that, the amylose content of test samples was 11%-15%,and the amylopectin content was 45%-65% from regressive equations;the results of Accuracy-Experiments and Standard-Addition-Experiments showed that the RSD of determination results of 5 times repeated experiments was less than 1 and the recovery was in 95.46%-102.23%, which indicated that stability and repeatability of the method was good enough to determine the content of amylose and amylopectin in millet;the correlation analysis showed that male parent had genetic effect on the starch distribution of hydrid millet.
