摘要
目的:研究丹皮酚脂质体中丹皮酚、磷脂和胆固醇三者分子间相互作用。方法:采用示差扫描量热(DSC)和同步辐射X光衍射(XRD)两种方法研究了丹皮酚脂质体中丹皮酚、胆固醇和二棕榈酰磷脂酰胆碱(DPPC)分子间的相互作用。结果:DSC实验结果表明,4%的丹皮酚或者胆固醇都使DPPC脂质体的前相变消失,主相变温度从42℃降低到38℃,二者都使DPPC脂质体主相变的协同性明显下降,但丹皮酚使DPPC脂质体主相变协同性下降更多。同步辐射X光衍射峰的峰形分析表明,4%的丹皮酚或者胆固醇都使脂质体发生明显的分相现象,且丹皮酚诱导的分相现象更加明显,与DSC实验结果一致。结论:DSC和XRD实验结果都表明,丹皮酚脂质体中胆固醇与DPPC疏水链的分子间相互作用要大于丹皮酚与DPPC疏水链的分子间相互作用,预示着在确定丹皮酚脂质体的处方时胆固醇的浓度不宜过大,否则将影响丹皮酚的包封率。
Objective:To investigate molecular interaction among paeonol, cholesterol and phospholipid in pae-onol-encapsulated liposomes.Methods:Differential scanning calorimetry ( DSC) and synchrotron x-ray dif-fraction ( XRD) techniques were employed to investigate molecular interaction among paeonol, cholesterol and dipalmitoyl-phosphatidylcholine ( DPPC) in paeonol-encapsulated liposomes.Results:DSC data show that the pretransition of DPPC liposomes containing 4 %paeonol or cholesterol disappeared and the temperature of main transition decreased from 42℃to 38℃.Though the cooperativity of main transition of DPPC liposomes containing 4 %paeonol or cholesterol both decreased obviously, it was paeonol that caused a smaller cooperat-ivity.XRD data showed that phase separation appeared in the DPPC liposomes containing 4%paeonol or cho-lesterol and it was paeonol that caused a more obvious phase separation, which was in accordance with DSC data.Conclusion:Both DSC data and XRD data show that the molecular interaction between cholesterol and hydrocarbon chains of DPPC is stronger than that between paeonol and hydrocarbon chains of DPPC.This re-sult suggests that the high concentration of cholesterol in paeonol-encapsulated liposomes can suppress the entrapment efficiency of paeonol.
