摘要
目的通过酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA)检测增殖性糖尿病视网膜病变(proliferative diabetic retinopathy,PDR)患者血清中的Dickkopf-1(DKK1)蛋白浓度,探讨Wnt信号通路在PDR发病中的作用。方法纳入2014年6月至12月于北京安贞医院及北京同仁医院眼科接受玻璃体视网膜手术的28例PDR患者及25例年龄、性别相匹配的非糖尿病对照组。通过ELISA法检测PDR组和对照组、活跃期和静止期PDR组血清中的DKK1蛋白浓度,分别比较组间的DKK1水平差异。结果 PDR组血清DKK1蛋白浓度为96.14~1303.65 pg·mL-1,平均为491.79 pg·mL-1;对照组血清DKK1蛋白浓度为250.36~1520.92 pg·mL-1,平均为882.38 pg·mL-1;两组相比,差异有统计学意义(P=0.002)。静止期PDR组血清DKK1蛋白平均浓度为683.35 pg·mL-1,高于活跃期PDR组(275.48 pg·mL-1),差异有统计学意义(P=0.022)。结论 PDR患者血清中DKK1蛋白水平明显降低,提示Wnt信号通路可能在PDR病理过程中发挥作用。
Objective To examine Dickkopf-1 (DKK1) levels in the serum of pa- tients with proliferative diabetic retinopathy (PDR) by enzyme-linked immunosorbent assay (ELISA) ,and evaluate the role of Wnt signaling pathway in the development of PDR. Methods Twenty-eight cases with PDR received pars plana vitrectomy in Bei- jing Anzhen Hospital and Beijing Tongren Hospital from June 2014 to December 2014 were prospectively recruited. Another 25 non-diabetic mellitus control cases were matched with age and gender. DKK1 levels in the serum of PDR group ( active PDR,qui- escent PDR) and controls were measured by ELISA. Mann-Whitney U test was used to analyze the differences between two groups. Results Serum levels of DKK1 in PDR group was 491.79 pg · mL-1 (96.14 - 1303.65)pg · mL-1 ,which in non-diabetic mellitus group was 882.38 pg · mL-1 (250.36 - 1520. 92)pg · mL-1 ,the PDR group was lower than the non-diabetic mellitus group (P = 0. 002 ). Furthermore, the serum concentra- tions of DKK1 in quiescent PDR group (683.35 pg · mL-1 ) was higher than that in ac- tive PDR group (275.48pg· mL-1)(P=0.022).Conelusion DKKI levels in the ser- um of patients with PDR are significantly decreased,indicating the participation of Wnt signaling pathway in the pathological processes of PDR.
