摘要
目的观察红花黄色素对乳腺癌耐药细胞MCF-7/ADR缝隙连接蛋白Cx43的影响。方法取对数生长期的MCF-7/ADR细胞,以2×105cells/孔接种于6孔培养板,24 h后分为4组,每组3孔,其中3组分别加入红花黄色素至终浓度25、50、100 mg/L,另一孔设为空白对照组,作用48 h。Western blot法检测各组MCF-7/ADR细胞中Cx43表达水平。结果不同浓度红花黄色素作用于MCF-7/ADR细胞48 h后,Cx43表达水平均增高,有明显的剂量依赖关系,以50、100 mg/L浓度作用显著(P<0.01)。结论红花黄色素能提高乳腺癌耐药细胞MCF-7/ADR缝隙连接蛋白Cx43的表达水平。
Objective To observe the effects of Safflower yellow on gap junction protein Cx 43 in drug resist-ant breast cancer cell line MCF -7/ADR.Methods MCF-7/ADR cells of logarithmic growth phase were inocula-ted in six-well culture dishes (2 ×10^5 cells each hole),and divided into 4 groups after 24 hours,3 holes in each group.3 groups were added Safflower yellow to the final concentration of 25,50,100 mg/L,respectively.The last group was blank control group.48 h after cultivating,the expression level of Cx43 in drug resistant breast cancer cell line MCF-7/ADR were detected by Western -blot.Results 48 h after different concentration of Safflower yellow acted on MCF-7/ADR cells,the expression level of Cx 43 was increased in a dose -dependent relationship and there were remarkable effects on the concentration of 50,100 mg/L (P〈0.01).Conclusion Safflower yellow can improve the expression level of gap junction protein Cx 43 in drug resistant breast cancer cell line MCF -7/ADR.
