摘要
【目的】探讨姜黄素对急性心肌缺血-再灌注损伤大鼠内皮素-1的影响。【方法】SD大鼠48只,随机分6组:假手术组(C)、缺血组(I)、缺血-再灌注1h组(R1h)、缺血-再灌注2h组(R2h)、姜黄素+缺血-再灌注1h组(Cur+I30min+R1h)和姜黄素+缺血-再灌注2h组(Cur+I30min+R2h)。酶联免疫法测量内皮素(ET-1)血清学水平;RT-PCR、Western blotting检测(MI/R)各组心肌组织中ET-1的变化,及Cur对ET-1表达的影响。【结果】与C组比较,I、R组血清ET-1明显升高(P<0.05);与I组比较,R组血清ET-1升高显著(P<0.05);Cur+R1、2h组血清ET-1较R1、2h明显下降(P<0.05)。与C组比较,I组ET-1mRNA表达无差异,ET-1蛋白水平升高(P<0.05);R组ET-1mRNA和蛋白表达随时间延长逐渐升高(P<0.05);Cur+R1、2h组ET-1mRNA和蛋白均较R1、2h组下降(P<0.05)。【结论】心肌缺血-再灌注损伤可试用姜黄素进行干预降低ET-1的表达。
[ Objective ] To investigate the effects of curcumin (Cur) on endothelin-1 (ET-1) in acute myocardial ischemia-reperfusion (MI/R) injury rats. [ Methods] Forty-eight SD rats were randomized into 6 groups: sham group (C), isehemia 30 minutes group (I), isch- emia 30 minutes + reperfusion 1 hour group (Rlh), ischemia 30 minutes + reperfusion 2 hours group (R2h), Curcumin + ischemia 30 minutes + reperfusion 1 hour group (Cur+I 30 min+Rlh), Curcumin + ischemia 30 minutes + reperfusion 2 hours group (Cur+I30min+ R2h). The levels of serum ET-1 were determined by ELISA and the changes of myocardial ET-1 were measured by RT-PCR and West- ern blotting methods. [Results]Compared with C group, serum ET-1 of I and R groups rose obviously (P〈0.05). Compared with I group, serum ET-1 of R group rose significantly (P〈0.05). Compared with Rlh and R2h groups, serum ET-1 of Cur+I 30 min+Rlh and Cur+I 30 min+R2h groups decreased significantly (P〈0.05). Compared with C group, ET-1 mRNA expression of I group showed no sig- nificant difference, while ET-1 protein level increased (P〈0.05). ET-1 mRNA and protein expressions of R group increased gradually as the time was prolonged(P〈0.05). ET-1 mRNA and protein expressions of Cur+I 30 min+R l h and Cur+I 30 min+R2h groups were lower than those of R l h and R2h groups (P〈0.05). [ Conclusion ] Curcumin can be tried to reduce ET-1 expression in myocardial ischemia- reperfusion injury..
