摘要 胰岛因子1(ISL1)是重要的转录因子,在胚胎发育期广泛分布于全身多种组织细胞,而在成体组织中主要表达于胰岛和神经组织。在胰腺中,ISL1主要调控胰岛素、胰高血糖素、生长抑素、胰多肽等内分泌激素的表达,在胚胎期促进胰腺背侧间充质细胞和胰岛内分泌细胞的分化、发育、成熟,在出生后通过抑制细胞凋亡、促进细胞增殖来维持胰岛β细胞数量的稳态。ISL1通过与基因启动子上的特定元件TAAT/ATTA结合,并与其它蛋白质相互作用,实现对下游靶基因的转录调控,以完成多种生理功能。 Isletl (ISL1) is a key transcription factor widely distributed in multiple tissues in embryonic stage, whereas it is mainly expressed in pancreatic islets and nervous system in postnatal stage. In pancreas, ISL1 regulates the expression of endocrine hormones, such as insulin, glucagon, somatostatin and amylin. It is responsible for the development of dorsal pancreas mesenchyme and controls cell fate specification of four endocrine islet cell lineages in embryonic pancreas. While in adult pancreatic islets, it inhibits apoptosis and meanwhile promotes proliferation of islet 13 cells, consequently maintaining the homeostasis of cell number. By interacting with other transcription factors and binding with specific TAAT/ATTA elements on gene pro- moters, ISL 1 controls the transcription of downstream target genes for its physiological functions.
