摘要
提取金氏真蛇尾皂苷并研究其抗氧化和抑菌活性,为蛇尾纲动物资源的研究和开发利用提供理论依据。以金氏真蛇尾为原材料,通过乙醇提取、石油醚脱脂、正丁醇萃取及大孔树脂AB-8除杂,制得蛇尾皂苷,其熔点为241—256℃;通过羟自由基法和邻苯三酚自氧化法来测定不同浓度的皂苷溶液对不同自由基的清除能力,采用滤纸片法及最低抑菌浓度(MIC)法测定金氏真蛇尾皂苷对大肠杆菌、金黄色葡萄球菌、啤酒酵母菌和青霉的抑制作用。金氏真蛇尾皂苷对·OH和O2-具有较好的清除作用,对大肠杆菌、金黄色葡萄球菌和啤酒酵母菌具有较好的抑制作用,对青霉的抑制作用较弱。通过抗氧化和抑菌活性的研究,表明金氏真蛇尾皂苷具有较好的抗氧化及抑菌活性。
To extract ophiurasaponin from Ophiura kinbergi and analyze its antioxidant and antibacterial activity, and provide the scientific basis for the development and utilization of the Ophiuroidea resources. With Ophiura kinbergi as raw materials, ophiurasaponin of Ophiura kinbergi were obtained by extraction by ethanol, degrasing by petroleum ether, extraction by n-butanol and removing impurity by AB-8 macroporous resin. The ophiurasaponin's melting point oranged from 241-256 ~. Through the hydroxyl free radical and the pyrogallol oxidation method to determine the ophiurasaponin solution of different concentration of different free radical scavenging activity, antibacterial activity by filter paper method and minimum inhibitory concentration (MIC) method for the determination of ophiurasaponin from Ophiura kinbergi on Escherichia coli, Staphylococcus aureus, Saccharomyces cerevisiae Hansen and Penicillium glaucum. The ophiurasaponin from Ophiura kinbergi has better scavenging effect on .OH and 02, has good inhibitory effect on Escherichia coli, Staphylococcus aureus and Saccharomyces cerevisiae Hansen, weak inhibitory effect on Penicillium glaucum. Through the study of antioxidant and antibacterialactivity, the results showed that the ophiurasaponin from Ophiura kinbergi has preferable antioxidation and antibacterial activity.
