心康冲剂对慢性心衰大鼠AVP mRNA表达的影响 被引量:5

摘要 目的:观察心康冲剂对慢性心衰大鼠AVP m RNA表达的影响。方法:70只SD大鼠随机抽取10只为正常对照组,剩余大鼠腹腔注射盐酸多柔比星建立CHF模型,再随机分为模型对照组、心康冲剂低中高剂量组、芪苈强心组(对照组),每组各12只。给药治疗第6周,各组随机取存活大鼠6只,开始收集大鼠24 h尿液(1次/d,连续7 d),实验结束后检测大鼠下丘脑AVP m RNA的表达情况。结果:与正常对照组比较,模型组的AVP m RNA表达显著升高,24 h尿量明显减少,差异有统计学意义(P<0.05),表明造模成功;与模型组比较,芪苈强心组、心康冲剂各组AVP m RNA表达显著下降,各组尿量均显著增加(P<0.05);与芪苈强心组比较,心康冲剂各组的AVP m RNA表达差异显著,各组尿量均显著增加(P<0.05);心康冲剂各组间比较AVP m RNA表达及尿量差异无统计学意义(P>0.05)。结论:心康冲剂通过下调心衰大鼠AVP m RNA表达,抑制下丘脑释放AVP从而减少水重吸收,增加尿量,可能系其对慢性心衰利尿消肿的作用机制之一。 Objective:To observe the effects of Xinkang granule on the AVP mRNA expression of chronic heart failure (CHF) rats hypothalamus. Method: The 10 rats in normal control group were randomly selected from 70 SD rats, the remaining rats by intraperitoneal injection of doxorubicin established CHF model, and then randomly divided into model group, low-, mid- dle- and high-dose Xinkang Granules groups, Qiliqiangxin group (control group ), 12 in each group. After 6 weeks of treatment administered rats, 6 rats were randomly selected from alived rats every group, 24-hour urine were collected (1 times/d, continu- ous 7d), and the expression of rat hypothalamus AVPmRNA was tested end of the experiment. Results: Model control group compared with normal control group, 24-hour urine decreased and AVP mRNA expression increased, there were significant dif- ference (P〈0.05), showed that the modeling was success. Compared with the model group, AVP mRNA expression decreased and the urine volume increased in Qiliqiangxin group and all Xinkang Granules groups (P〈0.05). Compared with Qiliqiangxin group, the AVP mRNA expression of all Xinkang Granules groups significantly decreased, 24-hour urine increased (P〈0.05); no signifi- cant difference between Xinkang granules groups (P〉0.05). Conclusion: Xinkang Granules can downregulat the expression of AVP mRNA, inhibit the release of AVP in the hypothalamus to reduce water reabsorption, and increase urine output, which is proba- bly one of its mechanism of treat water retention of chronic heart failure.
