南疆43份引进及本地苜蓿种质资源同工酶分析 被引量:1

Analysis of 43 Introduced and Local Alfalfa Germplasm Resources of Southern Xinjiang Using Isozyme Markers

摘要 【目的】对南疆地区引进及本地苜蓿种质资源的亲缘关系进行分析,为地区苜蓿新品种培育提供科学依据。【方法】以43份引进苜蓿品种、新疆大叶和拜城野生紫花苜蓿为材料,用过氧化物酶、淀粉酶、过氧化氢酶和酯酶电泳技术对其亲缘关系进行分析。【结果】4种同工酶在43份苜蓿材料中共检测到38条稳定酶带,过氧化物酶、淀粉酶、过氧化氢酶和酯酶的多态位点百分率分别为91.67%、66.67%、66.67%和75.00%。材料间的相似系数0.579-0.974,平均为0.798。43份材料在相似系数0.725处可聚为3类,拜城野生紫花苜蓿和河西紫花苜蓿分别被单独聚为类。捷克、拜城野生紫花苜蓿、河西、赛特、格林、渭南、秘鲁与大部分材料的亲缘关系较远。【结论】43份苜蓿材料具有一定水平的遗传差异,部分材料具有遗传特异性。 【Objective】The purpose of this study is to investigate the genetic relationship of introduced and local alfalfa germplasm resources of Southern Xinjiang and provide scientific basis for future alfalfa breeding programs.【Method】The genetic relationship of 41 introduced alfalfa cultivars,Xinjiangdaye and Baicheng wild alfalfa were analyzed by using peroxidase,amylase,catalase and esterase electrophoresis.【Result】The results showed that 4 isozymes generated a total of 38 stable bands across 43 alfalfa accessions,and the percentage of polymorphic bands for peroxidase,amylase,catalase and esterase were 91. 67%,66. 67%,66. 67% and75. 00%,respectively. The similarity coefficient among alfalfa accessions ranged from 0. 579 to 0. 974 with an average of 0. 798. The accessions could be classified into 3 groups at the similarity coefficient value of 0. 725 based on cluster analysis,among which baicheng wild alfalfa and hexi were clustered into 2 independent groups. According to principal component analysis,czech,baicheng wild alfalfa,hexi,saite,grimm,weinan andperu had relatively distant genetic relationships with most accessions.【Conclusion】There were a certain genetic differences among 43 alfalfa accessions,and some accessions had genetic specificity.
