摘要 建立检测抗疲劳药物及保健品中非法添加的8种壮阳类化学药物的方法。采用高效液相色谱~质谱串联(HPLC—MS/MS)法,watersAtiantis dCl8型色谱柱(150mm×2.1mm×3μm),乙腈-0.1%甲酸溶液为流动相,进行梯度洗脱,选择正离子多反应监测扫描方式检测。结果表明,硫代豪莫西地那非、硫代西地那非、羟基红地那非、育亨宾、羟基伐地那非、二甲基西地那非、去甲基西地那非、乌地那非的检测质量浓度线性范围分别为96—9600ng/mL(r=-0.9991),86—8600ng/mL(r=0.9987),97~9700ng/mL(r=0.9990),102~10200ng/mL(r=0.9979),101-10100ng/mL(r=0.9983),102~10200ng/mL(r=-0.9995),93-9300ng/mL(r=-0.9988),89-8900ng/mL(1=-0.9992),检测限分别为44,17,33,56,36,92,26,51ng/mL,平均回收率分别为97.6%,94.5%,90.2%,98.8%,99.6%,9513%,103.6%,96.4%(RSD均〈5%,n=6)。结果表明,该方法能够快速、准确地检测出抗疲劳药物及保健品中非法添加的8种壮阳类化学药物。 To establish a method for the determination of 8 illegal added anti-impotence medicines in anti-fatigue preparations and health care products. HPLC-MS/MS method is adopted. The determination is performed on waters Atiantis dG18 (150 mm× 2.1 mm × 3 μm) column, the mobile phase is acetonitrile - 0.1% formic acid with Gradient elution, under positve ion multiple reaction monitoring mode. Results show that: the linear ranges are 96-9 600 ng/mL for hydroxyhomosildenafil (r= 0.999 1) , 86-8 600 ng/mL for thiosildenafil (r=0.998 7) , 97-9 700 ng/mL for hydroxyacetildenafil (r=0.999 0) , 102- 10 200 ng/mL for yuhimbine (r=-0.997 9), 101~10 100 ng/mL for hydroxyvardenafil (r=-0.998 3), 102~10 200 ng/mL for dimethylsildenafil (r=-0.999 5), 93~9 300 ng/mL for desmethyl sildenafil (r=-0.998 8) and 89-8 900 ng/mL for udenafil (r=-0.999 2) . The lowest limits of detection are 44, 17, 33, 56, 36, 92, 26, 51 ng/mL respectively. Average recoveries are 97.6%, 94.5%, 90.2%, 98.8%, 99.6%, 95.3%, 103.6%, 96.4% (RSD〈5%, n=6) . The method is quick and accurate for the determination of 8 illegal added anti-impotence medicines in anti-fatigue preparations and health care products.
