河南汉族人群MTHFR基因多态性及同型半胱氨酸与冠心病关系研究 被引量:10

Association of polymorphism of 5,10-methylenetetrahydrofolate reductase and plasma homocysteine with coronary heart disease in Henan Han population

摘要 目的探讨河南地区汉族人群5,10-亚甲基四氢叶酸还原酶(5,10-methylenetetrahydrof01atereductase,MTHFR)C677T多态性及血清同型半胱氨酸水平与冠心病的相关性。方法河南汉族308例冠心病患者(冠心病组)和296例体检健康者(对照组),2组采用全自动生化分析仪检测血清同型半胱氨酸(homocysteine,Hcy)水平,采用PCR-测序技术检测MTHFR基因型。结果冠心病组血清Hcy水平[(16.98±10.16)/μmol/L]明显高于对照组[(11.87±7.69)μmol/L](P〈0.05);冠心病组和对照组TT基因型血清Hcy水平[(21.03±10.37)、(15.07±4.36)μmol/L]明显高于CT型[(16.58±8.87)、(12.86±4.37)μmol/L]及CC型[(13.16±6.97)、(10.18±4.83)μmol/L](P〈0.05),CT型明显高于CC型(P〈O.05);冠心病组MTHFRC677TTT基因型频率(32.8%)明显高于对照组(18.6%),CC基因型频率(19.5%)明显低于对照组(36.8%)(P〈O.05);携带TT基因型是患冠心病的独立危险因素(OR=2.138,95%CI:1.466~3.118,P=0.000)。结论河南汉族人群MTHFR基因多态性及Hcy水平与冠心病的发生相关。 Objective To investigate the correlation of the polymorphism of 5,10-methylenetetrahydrofolate reductase(MTHFR)C677T and serum homocysteine(Hcy)with coronary heart disease(CHD)in Henan Han population.Methods The Hcy level was detected automatic biochemical analyzer,and genotypes of MTHFR was detected by PCRsequencing in 308 Han patients with CHD(CHD group)and 296 Han healthy volunteers(control group)in Henan.Results The Hcy level was higher in CHD group((16.98±10.16)μmol/L)than that in control group((11.87±7.69)μmol/L)(p〈0.05).In CHD group and control group,the Hcy levels of TT genotype((21.03±10.37),(15.07±4.36)μmol/L were the highest,followed by CT genotype((16.58±8.87),(12.86±4.37)μmol/L)and CC genotype((13.16±6.97),(10.18±4.83)μmol/L)in turns(p〈0.05).The TT genotype frequency of MTHFR C677 T was significantly higher and the CC genotype frequency was significant lower in CHD group(32.8%,19.5%)than those in control group(18.6%,36.8%)(p〈0.05).The TT genotype of MTHFR C677 T was an independent risk factor of CHD(OR=2.138,95%CI:1.466 to 3.118,P=0.000).Conclusion The polymorphism of MTHFR C677 Tand serum Hcy level are involved in CHD in Henan Han population.
