红花中羟基红花黄色素A的提取工艺 被引量:4

摘要 采用单因素实验和4因素3水平正交实验,利用面积归一法,以高效液相检测羟基红花黄色素A的含量为指标,考察了羟基红花黄色素A提取工艺的影响因素,确定了最佳的溶剂浓度、溶剂用量、提取温度和超声时间.结果表明超声提取时间对羟基红花黄色素A提取的影响最为显著,影响因素的主次顺序为:超声时间﹥乙醇浓度﹥提取温度﹥溶剂用量.最佳单因素为:70%的乙醇质量浓度、50℃的提取温度、体积比为15的溶剂用量、15 min的超声时间;最佳提取工艺为:在50℃下,加入10倍量的质量分数70%乙醇,超声提取20 min,固定提取次数为一次,此时羟基红花黄色素A的提取含量最高. According to the content of hydroxy safflower yellow A(HSYA) by high performance liquid chromatography, single factor experiments and orthogonal experiments(four factors three levels) were conducted to determine the optimal factors on the extraction of HSYA, such as the concentration of solvent, the amount of solvent, extraction temperature and extraction time. The results show that the ultrasonic extraction time is the most important factor and the order of the influential factors are ultrasonic extraction time〉ethanol concentration〉extraction temperature 〉solvent consumption. The conditions for the best single factor experiment are 70% ethanol concentration, 15 times of the volume ratio of solvent and ultrasonic extract for 15 minutes under 50 ℃. The best extraction factor for the highest content of HSYA is adding 10 times of 70% ethanol extract only once for 20 minutes under 50 ℃ with ultrasonic.
