摘要
目的:建立HPLC测定梅花配方颗粒中芦丁、金丝桃苷、异槲皮苷和槲皮素含量的方法。方法:Phenomenex Synergi C18色谱柱(4.6 mm×250 mm,5μm),流动相乙腈(A)-0.1%磷酸(B)梯度洗脱,柱温30℃,体积流量1.0 m L·min-1,检测波长360 nm。结果:芦丁、金丝桃苷、异槲皮苷和槲皮素线性范围分别为0.230 2~2.877 5μg(r=0.999 2),0.134 8~1.685 0μg(r=0.999 5),0.195 4~2.442 5μg(r=0.999 1),0.033 8~0.422 5μg(r=0.999 9)线性关系良好,平均加样回收率分别为98.39%,98.71%,100.00%,100.57%;RSD分别为1.7%,1.4%,1.8%,2.0%。结论:该测定方法简便快捷、准确、重复性好,为梅花配方颗粒的质量控制和评价提供了依据。
Objective: To establish an HPLC method for simultaneous determination ofrutin, hyperin, isoquercitrinand quercetinin Mone Flose dispensing granule. Method: HPLC method was performed on a Phenomenex C18 (4.6 mm× 250 ram, 5 p,m) with Acetonitrile (A) -0. 1% phosphoric acid (B) as mobile phase by gradient elution. The flow rate was 1.0 mL ·min, column temperature was 30 ℃ and detection wavelength was 360 nm. Result: Rutin, hyperin, isoquercitrinand quercetin showed good linearity in the ranges of 0.230 2-2.877 5 μg (r=0.9992), 0.1348-1.6850 μg (r=0.9995), 0.1954-2.4425 μg (r=0.999 1) and 0.033 8-0. 422 5 μg (r = 0.999 9) with average recoveries of 98.39%, 98.71%, 100.00% and 100.57% , with RSD 1.7% , 1.4% , 1.8% and 2.0% , respectively. Conclusion: The method is rapid, accurate and repeatable, which can provide a reliable way for evaluating the quality of Mone Flose dispensing granule.
