摘要
目的 建立高效液相色谱法同时测定不同产地凤尾草中的蕨素C-3-O-葡萄糖苷、忍冬苷和野漆树苷3种活性成分的含有量。方法 凤尾草甲醇提取液分析采用Diamonsil C18色谱柱(4.6 mm×250 mm,5μm);流动相为乙腈-0.15%甲酸水溶液,梯度洗脱;体积流量1.0 m L/min;检测波长258 nm和350 nm;柱温30℃。结果 蕨素C-3-O-葡萄糖苷、忍冬苷和野漆树苷3种活性成分分别在0.025 0~1.60、0.054 0~3.45和0.238~15.2μg的测定范围内表现出良好的线性关系(r〉0.999),加样回收率分别为101.0%(RSD 1.4%)、101.0%(RSD 1.8%)和101.6%(RSD1.2%)。10个不同来源凤尾草中的蕨素C-3-O-葡萄糖苷最高与最低含有量相差约15倍,忍冬苷含有量相差约8倍,而野漆树苷含有量相差达25倍。结论 该方法能同时测定凤尾草中3种活性成分的含有量,而且其药材质量与产地关系较大。
AIM To establish an HPLC method for simultaneously determining three constituents( pterosin C3-O-β-D-glucosidede,lonicerin and rhoifolin) in Pteris multifida from different growing areas. METHODS The P. multifida methanol extract was carried out on a Diamonsil C18Column( 4. 6 mm × 250 mm,5 μm),the mobile phase was acetonitrile-water( containing 0. 15% formic acid) in a gradient elution manner,the flow rate was 1. 0m L / min at 30℃,and the detection wavelength was set at 258 nm and 350 nm,respectively. RESULTS Excellent chromatographic separation was achieved with good linearity( r 0. 999) within the studied concerntration ranges. The recovery ranges for pterosin C 3-O-β-D-glucosidede, lonicerin and rhoifolin were 101. 0%( RSD1. 4%),101. 0%( RSD 1. 8%),and 101. 6%( RSD 1. 2%),respectively. The difference of pterosin C 3-O-β-D-glucosidede content in P. multifida collected from 10 growing areas between maximum and minimum value reached 15 times,of lonicerin about 8 times,and of rhoifolin as high as 25 times. CONCLUSION The method can simultaneously determine the 3 constituents in P. multifida,and the growing areas of this plant are closely linked to its quality.
