煤氟污染的粮食对动情期雌鼠全血铜、锌、铁、钙、镁元素的影响

Effect of coal fluoride contaminated food on copper,zinc,iron,calcium,and magnesium levels in whole blood of female rats during estrum

摘要 目的 了解食用煤氟污染的粮食对动情期雌鼠全血铜(Cu)、锌(Zn)、铁(Fe)、钙(Ca)、镁(Mg)元素的影响。方法 选取贵阳医学院动物实验中心提供的90只断乳2周、体重60-80 g的SD雌性大鼠为研究对象,随机分为对照组、中氟组、高氟组,每组各30只。各染氟组喂饲含不同比例的燃煤型氟中毒病区煤烘烤玉米(含氟量分别为47.8 mg/kg、96 mg/kg)复制燃煤型氟中毒模型,对照组玉米饲料来自非病区(含氟量为5.2 mg/kg)。分别于染氟60、120、180天时于股动脉取血,检测Cu、Zn、Fe、Ca、Mg元素水平。结果 60天时染氟组Cu、Zn、Fe明显降低,高氟组Ca明显增高,差异有统计学意义(P均〈0.05);3组Mg随染氟剂量增加而降低,差异无统计学意义(P〉0.05)。120天时染氟组Cu均升高;高氟组Zn高于对照组、中氟组Zn随染氟剂量增加而升高,差异有统计学意义(P〈0.05);120天时高氟组Fe降低,差异有统计学意义(P〈0.05)。180天时染氟组Cu、Zn、Fe、Ca、Mg接近对照组水平,差异无统计学意义(P〉0.05)。染氟组Cu在120、180天高于60天,差异有统计学意义(P〈0.05),且随染氟时间的延长逐渐升高。染氟组Zn在120、180天高于60天,差异有统计学意义(P〈0.05),高氟组Zn在180天低于120天,差异有统计学意义(P〈0.05)。染氟组Fe在120、180天高于60天,随着中毒时间延长逐渐升高,除中氟组120、180天之间差异无统计学意义外(P〉0.05),其余各组间比较差异均有统计学意义(P〈0.05)。中氟组Ca在120、180天高于60天,随染毒时间延长,全血Ca含量逐渐增加,仅在60、180之间差异有统计学意义(P〈0.05);高氟组60、120、180天全血Ca含量基本维持同一水平,差异无统计学意义(P〉0.05)。3组Mg均随时间延长而逐渐增加,差异有统计学意义(P〈0.05)。结论 燃煤型氟中毒可导致动情期雌鼠全血Cu、Zn、Fe、Ca、Mg水平紊乱,不同染氟剂量在不同染氟时间对这些元素影响错综复杂。 Objective:To understand the effect of coal fluoride contaminated food on copper,zinc,iron,calcium,and magnesium levels in whole blood of female rats during estrum.Methods:Ninety SD female rats(delectation for two weeks,60-80 g) provided by animal experimental center in Guiyang Medical College were selected and randomly divided into control group,moderate fluoride exposure group,and high fluoride exposure group,30 rats in each group.Rat model of coal-burning fluorosis was established by feeding the rats in moderate fluoride exposure group and high fluoride exposure group with baked corn(the contents of fluorine:47.8 mg / kg,96 mg / kg); the rats in control group were fed with normal corn from non-endemic areas(the content of fluorine:5.2 mg / kg).Femoral artery blood specimens were obtained at 60,120,and 180 days after fluoride exposure to detect the levels of copper,zinc,iron,calcium,and magnesium.Results:At 60 days after fluoride exposure,the levels of copper,zinc,and iron in two fluoride exposure groups decreased significantly,the level of calcium in high fluoride exposure group increased significantly,there were statistically significant differences(P〈0.05); the levels of magnesium in the three groups decreased significantly with the increase of fluoride content,but there was no statistically significant difference(P〈0.05).At 120 days after fluoride exposure,the levels of copper in the two fluoride exposure groups increased; the level of zinc in high fluoride exposure group was higher than those in control group and moderate fluoride exposure group,the levels of zinc increased significantly with the increase of fluoride content(P〈0.05); the level of iron in high fluoride exposure group decreased significantly(P〈0.05).At 180 days after fluoride exposure,there was no statistically significant difference in the levels of copper,zinc,iron,calcium,and magnesium between the two fluoride exposure groups and control group(P〈0.05).The levels of copper and zinc in the two fluoride exposure groups at 120 and 180 days after fluoride exposure were statistically significantly higher than those at 60 days after fluoride exposure(P〈0.05),which increased gradually with the exposure time to fluoride.In high fluoride exposure group,the level of zinc at 180 days after flu-oride exposure was statistically significantly lower than that at 120 days after fluoride exposure(P〈0.05).In the two fluoride exposure groups,the levels of iron at 120 and 180 days after fluoride exposure were higher than those at 60 days after fluoride exposure,which increased gradually with the exposure time to fluoride,there were statistically significant differences among different time points after fluoride exposure between the two fluoride exposure groups(P〈0.05),except between 120 days and 180 days in moderate fluoride exposure group(P〈0.05).In moderate fluoride exposure group,the levels of calci-um at 120 and 180 days after fluoride exposure were higher than those at 60 days after fluoride exposure,which increased gradually with the exposure time of fluoride,but there was only statistically significant difference between 60 days and 180 days(P〈0.05).In high fluoride exposure group,there was no statistically significant difference in the level of calcium among 60,120,and 180 days after fluoride exposure(P〈0.05).The levels of magnesium in the three groups increased gradually with the exposure time to fluoride,there were statistically significant differences(P〈0.05).Conclusion:Coal-burning fluorosis can induce disorder of copper,zinc,iron,calcium,and magnesium levels in whole blood of female rats during estrum,different fluoride exposure levels have different impacts on the levels of the above-mentioned elements at different time points.
