摘要
目的探讨结直肠癌组织中mi R-196b和Hox B8的表达及临床意义。方法选取2012年10月-2014年3月在该院手术治疗的47例结直肠癌患者为研究对象,术中留取结直肠癌组织及正常黏膜组织,应用实时荧光定量PCR技术检测二者mi R-196b m RNA和Hox B8 m RNA表达情况,Hox B8蛋白表达检测应用Western blot,对检测结果进行统计学分析。结果结直肠癌组织中mi R-196b m RNA和Hox B8 m RNA表达量显著高于正常黏膜组织(P<0.01),且结直肠癌中两者表达水平呈显著负相关(r=-0.462,P=0.012);mi R-196b表达与肿瘤分期及淋巴结转移相关(P<0.05),而Hox B8表达与结直肠癌临床病理特征不具有相关性(P>0.05)。结论结直肠癌组织中mi R-196b及Hox B8表达水平显著升高,mi R-196b不仅参与结直肠癌的发生发展,而且可能抑制Hox B8表达。
【Objective】To investigate the expression of mi R-196 b and Hox B8 in colorectal cancer tissues and its clinical significance.【Methods】Forty-seven cases of colorectal cancer who underwent surgical treatment from October 2012 to March 2014 in our hospital were selected as the research subjects. The colorectal cancer tissue and normal mucosa tissue of each patient were collected during the operation. Real-time fluorescence quantitative PCR technique was used to detect both the mi R-196 b m RNA and Hox B8 m RNA expressions. Hox B8 protein expression was analyzed by Western blot. Finally, the test results were statistically analyzed. 【Results】 The expressions of mi R-196 b m RNA and Hox B8 m RNA in the colorectal cancer tissues were significantly higher than those in the normal mucosa tissues(P 〈 0.01), and their expression levels in colorectal cancer were negatively correlated(r =-0.462,P = 0.012). The mi R-196 b expression was closely correlated with tumor stage and lymph node metastasis(P 〈 0.05),and Hox B8 expression had no correlation with the clinical pathological characteristics of colorectal cancer(P 〉 0.05).【Conclusions】The expression levels of mi R-196 b and Hox B8 are significantly increased in colorectal cancer tissues. Mi R-196 b not only participates in the development and progression of colorectal cancer, but also might inhibit the expression of Hox B8.
