摘要
目的:明确针刺内关穴对大脑中动脉缺血(MCAO)大鼠血管内皮细胞增殖的影响,探讨脑缺血后病理损伤和修复的可能机制。方法:复制大鼠MCAO模型,随机分为4组,模型组、假手术组、非针刺组、针刺组(内关穴3次/s、60s),每组6只模型。4组取材后,通过t-检验结合差异变化倍数(FC)筛选进行差异分析,检测差异表达基因、GO。结果:模型组与假手术组相比,脑缺血病理损伤致基因上调417个,共富集到340个GO生物过程;针刺组与非针刺组相比较,针刺组大鼠神经行为学评分有显著差异,肢体症状较非针刺组减轻(P≤0.05),下调358个基因,共富集到19个GO生物过程。结论:脑缺血损伤导致显著变化的生物过程包括单核细胞聚集、正向调控血管生成、急性时相反应、中性粒细胞趋化性、病毒防御应答、正向调控异型细胞的细胞黏附等,是一个复杂的生物过程;针刺内关穴治疗脑缺血的机制,可能与调控Egr3、Ccl11、Vash2等表达促进大鼠血管内皮细胞增殖有关,通过促进大鼠缺血脑组织脑血管新生,改善脑缺血症状。
Objective: To study the effects of acupuncture on positive regulation of endothelial cell proliferation in brain tissue of rats with cerebral ischemia,and to discuss the mechanism of tissue damage and repair. Methods: MCAO was created by thread embolus,which were randomly divided into four groups with 6 cases in each: model,so,no-acu,acu. The Gene and GO were studied and compared after acupuncture. Results: model VS so: there were 340 different expression GO BP of 417 up-regulated genes; acu VS no-acu : there were 19 different expression GO BP of 358 down-regulated genes. Conclusion: Acupuncture on Neiguan( PC6) can promote the recovery of positive regulation of endothelial cell proliferation in brain tissue of rats with cerebral ischemia.
