摘要
以‘李广杏’幼胚(盛花后30~40d)为试验材料,MS为基本培养基,研究了激素及其不同浓度组合对幼胚萌发和胚培苗继代增殖的影响.结果表明:幼胚萌发的适宜培养基配方为MS+6-BA 0.5 mg/L+IAA0.2mg/L,在培养20d时萌发率达到100%,子叶生长健壮,叶片浓绿伸展;胚培苗继代增殖的适宜培养基配方为MS+6-BA 1.0mg/L+IAA 0.05mg/L,增殖系数达3.42.
The experiment studied the effects of various plant growth regulators and their different concentration combinations on immature embryo germination and embryo seedlings subculture proliferation of apricot variety Liguangxing.Using young embryos(30dto 40 dafter full blooming)were used as experimental materials and MS was the basic medium.The result indicated that the immature embryo grew well on MS+6-BA 0.5mg/L+ IAA 0.2mg/L medium during its primary culture.Under this processing,the germination rate reached 100% at 20 th day,cotyledons grew strongly,leaves dark green and stretch.The suitable subculture medium of immature embryos is MS + 6-BA 1.0mg/L +IAA 0.05mg/L.Under this treatment,the propagation coefficient reached 3.42.The techniques of immature embryo culture of apricot variety‘Liguangxing'in vitro are established in this study,which lay a foundation for its cross breeding and genetic transformation.
