白藜芦醇对佐剂性关节炎大鼠血管内皮因子表达的影响 被引量:5

摘要 目的探讨白藜芦醇(Res)对佐剂性关节炎(AA)模型大鼠关节滑膜组织的抗炎作用和对血管内皮生长因子(VEGF)表达水平的影响。方法 60只SD大鼠随机分正常组(10只)和模型组(50只),模型组左后足趾皮内注射弗氏完全佐剂建立AA模型,模型组在建模后12 d再分5组:模型对照组、Res低剂量组(5 mg/kg)、Res中剂量组(15 mg/kg)、Res高剂量组(30 mg/kg)、阳性药塞来昔布组(5 mg/kg),给药16 d后处死大鼠,对各组大鼠踝关节滑膜组织行病理切片、HE染色和VEGF免疫组化染色。观察滑膜增生、血管翳形成、炎症渗出、关节软骨和骨破坏等情况。Western blot法检测VEGF蛋白的表达情况。结果 Res治疗组的关节病理学评分明显低于模型对照组,免疫组化染色以及Western blot结果显示,Res能降低滑膜组织VEGF表达水平。结论 Res有改善大鼠关节滑膜炎症、抑制滑膜组织VEGF表达的作用,这可能与Res治疗AA关节炎有关。 Objective To investigate the effect of anti-inflammatory and expression level of vascular endothelial growth factor( VEGF) of resveratrol ( Res) on synovial tissues in rats with adjuvant arthritis ( AA) . Methods 60 SD rats were randomly divided into two groups:normal group (10 rats) , model group (50 rats) . Model group was injected freund′s complete adjuvant induced arthritis, when the right paw was induced after 12 days and secondary immune-inflammatory feet were swollen,the model rats were randomly divided into 5 groups, and each group had 10 rats AA model,Res low dose group(5 mg/kg) , Res middle dose group(15 mg/kg) , Res high dose group(30 mg/kg),positive control celecoxib group (5 mg/kg) qd × 16. After induced inflammatory paw, the expressions of VEGF in synovial tissue and articular cartilage was detected by immunohistochemistry, VEGF protein content in synovial tissue were detected by Western blot. Results Res could inhibit model group rats pathological damage, compared with the AA model group, synovial tissue expression of VEGF was significantly low in the resveratrol-trea-ted groups and celecoxib group. Conclusion Res prevents the pathological changes of the joint degeneration, its mechanism may be invovled regulating the expression of VEGF in synovial tissue of AA rats.
