IGF-1和IGF-1R在无功能性垂体腺瘤中的表达及意义 被引量:3

Expression of insulin-like growth factor 1 and its receptor in human non-functioning pituitary adenomas: correlation with invasiveness and recurrence

摘要 目的探讨胰岛素样生长因子1(insulin-like growth factor 1,IGF-1)和胰岛素样生长因子1型受体(insulin-like growth factor 1 receptor,IGF-1R)与无功能性垂体腺瘤侵袭性和复发性的关系。方法60例无功能性垂体腺瘤分为侵袭性复发性组(16例)、侵袭性非复发性组(24例)和非侵袭性非复发性组(20例)。采用qRT-PCR检测IGF-1和IGF-1R的mRNA表达水平,采用免疫组织化学法和Western blot检测IGF-1、IGF-1R和其下游信号分子血管内皮生长因子(vascular endothelial growth factor,VEGF)的蛋白表达水平。结果 IGF-1和IGF-1R的mRNA表达水平及IGF-1、IGF-1R和VEGF的蛋白表达水平,侵袭性复发性组高于侵袭性非复发性组(P<0.05),侵袭性非复发性组高于非侵袭性非复发性组(P<0.05)。相关性分析结果显示,在侵袭性复发性、侵袭性非复发性无功能性垂体腺瘤组织中IGF-1和IGF-1R蛋白表达量呈正相关(r=0.85,r=0.80,P<0.05),IGF-1R和VEGF蛋白表达量呈正相关(r=0.74,r=0.70,P<0.05);在非侵袭性非复发性无功能性垂体腺瘤组织中IGF-1和IGF-1R蛋白表达量呈正相关(r=0.73,P<0.05),IGF-1R和VEGF蛋白表达量无明显相关性(P>0.05)。结论IGF-1、IGF-1R和VEGF在侵袭性和复发性无功能性垂体腺瘤中表达上调,三者协调作用,与无功能性垂体腺瘤的侵袭性和复发性恶性行为的发生、发展可能相关。 Objective To investigate the expression levels of insulin-like growth factor 1( IGF-1) and its receptor( IGF-1R) in human non-functioning pituitary adenomas and their relationships with tumor invasion and recurrence. Methods Sixty specimens of non-functioning pituitary adenomas collected in surgery in our department from August 2012 to May 2014 were classified into invasive recurrent group( n = 16),invasive nonrecurrent group( n = 24) and non-invasive non-recurrent invasive group( n = 20) according to the clinical data of the corresponding patients. Quantitative real-time polymerase chain reaction( qRT-PCR) was used to detect the mRNA expression levels of IGF-1 and IGF-1R. Western blot assay and immunohistochemical assay were employed to measure the protein levels of IGF-1,IGF-1R and its downstream molecule of associated signaling pathway,VEGF. Results The mRNA expression levels of IGF-1 and IGF-1R and the protein expression levels of IGF-1,IGF-1R and VEGF were significantly higher in invasive recurrent groups than in invasive nonrecurrent group( P〈0. 05),and in the invasive non-recurrent group than non-invasive non-recurrent group( P〈0. 05). The correlation analysis revealed that there was a positive correlation between the protein levels of IGF-1and IGF-1R( r = 0. 85,r = 0. 80,P〈0. 05),and between those of IGF-1R and VEGF( r = 0. 74,r =0. 70,P〈0. 05) in invasive recurrent group and invasive non-recurrent group respectively. Positive correlation was also found between the protein levels of IGF-1 with IGF-1R in non-invasive non-recurrent group( r = 0. 73,P〈0. 05),but no obvious relation was found in the levels of IGF-1R with VEGF( P〈0. 05). Conclusion The expression of IGF-1,IGF-1R and VEGF is enhanced in the invasiveness and recurrence of non-functioning pituitary adenomas. The 3 molecules may play synergetic roles and are involved in the occurrence and development of invasive growth and recurrence of the tumor.
