摘要
目的对1例术前备血ABO血型血清学鉴定疑为Cis AB型的血样进行分子确认。方法应用单克隆抗体和人源抗体检测该患者红细胞ABO血型抗原,用ABO标准红细胞检测其血清中的ABO抗体,采用序列特异性聚合酶链反应(PCR-SSP)技术进行ABO基因的初步分型检测,并对ABO基因第6和第7外显子的核苷酸序列采用聚合酶链反应进行扩增,并对扩增产物进行纯化,然后进行测序反应,测序结果运用相关软件进行分析,最后将分析结果与红细胞血型基因突变数据库进行比对。结果血型血清学鉴定为Cis AB亚型,PCR-SSP技术基因分型为A/O型;ABO基因第6、第7外显子测序结果经与A101核苷酸序列比对发现如下杂合位点:261A/G,467T/C,803C/G。结论血型血清学对Cis AB和AB亚型判定有一定的局限性,分子手段可明确该样本的血型为cis AB型,基因型为Cis AB01/O01。
Objective The suspected Cis AB blood sample identified with ABO blood group serology was performed for molecular recognition. Methods The red blood cells ABO group antigens of the patient were identified by monoclonal antibodies and polyclonal antibodies respectively,and the ABO antibodies in serum were tested using standard ABO cells,the preliminary ABO genotype was evaluated with PCR- SSP technology,and then the exon 6,exon 7 of ABO gene were amplified by polymerase chain reaction( PCR) technique,which were purified and sequenced using the sequencing technology,respectively,the results were analyzed by special software and compared with the Blood Group Antigen Gene Mutation Database. Results The serological identification result was Cis AB subtype,PCR- SSP indicated the preliminary genotyping A / O,ABO gene exon 6,exon7 sequencing results by comparison with nucleotide sequences of A101 presented 261 A / G,467 T / C and 803 C / G. Conclusion There are,for serological methods,certain limits in identifying Cis AB and AB serotypes,which can be confirmed as CisAB by molecular means. The gene type is Cis AB01 / O01.
