摘要
目的研究人乳头瘤病毒(HPV)16型E6蛋白(HPV16-E6蛋白)与P73蛋白在宫颈鳞癌组织中的表达及两者的相关性,探讨HPV16-E6蛋白、P73蛋白在宫颈肿瘤发生、发展过程中的作用。方法选取正常宫颈组织10例、宫颈上皮内瘤样病变(CIN)组织22例、宫颈鳞癌组织38例,采用PV-6000二步法检测3种组织中HPV16-E6、P73蛋白的表达情况,分析两者与相关临床病理参数间的关系及两者之间的相关性。结果鳞癌组HPV16-E6蛋白的表达明显高于正常组、CIN组,差异有统计学意义(χ2=21.703,P<0.05)。鳞癌组P73蛋白表达明显低于正常组、CIN组,差异有统计学意义(χ2=15.461,P<0.05)。宫颈鳞癌组织中HPV16-E6蛋白阳性表达率与组织学分级显著相关(χ2=9.011,P<0.05);与发病年龄、临床分期、淋巴结转移及肿瘤大小无关(P>0.05)。HPV16-E6蛋白与P73蛋白的阳性表达呈负相关(rs=-0.301,P<0.05)。结论 HPV16-E6蛋白表达升高可能与宫颈鳞癌的发生、发展相关。随着宫颈肿瘤发展程度的增高,P73蛋白的表达逐渐降低,提示p73基因可能是宫颈肿瘤的抑癌基因。
Objective To investigate the expressions of human papillomavirus(HPV)16-E6 protein and P73 in tissue of squamous cell carcinoma(SCC)of the cervix and their correlation,and explore their role in the occurence and development of the disease. Methods Tissue of normal cervix was selected from 10 cases,that of cervical intraepithelial neoplasia(CIN)from 22 cases and that of SCC from 38 cases.Employing PowerVision two-step immunohistochemical method to detect the expressions of HPV16-E6 protein and P73 in the three kinds of tissue,and analyze the relationship between both HPV16-E6 protein and P73 and correlated clinical pathologic parameters,as well as the relationship between HPV16-E6 protein and P73. Results The positive rate of HPV16E6 protein in SCC was higher than that in normal cervix and CIN,the difference was significant(χ^2=21.703,P〈0.05).In SCC group,the expression of P73 was lower than that in normal and CIN groups,the difference was significant(χ^2=15.461,P〈0.05).In SCC group,the expression of HPV16-E6 protein was related to histological grade(χ^2=9.011,P〈0.05),and unrelated to the age of onset,clinical stage,lymph node metastasis and the size of tumor(P〉0.05).The expression of HPV16E6 was negatively correlated with that of P73(rs=-0.301,P〈0.05). Conclusion The elevation of HPV16-E6 expression is probably correlated with the occurence and progression of squamous cell carcinoma of the cervix.The expression of P73 gradually decreases with the progression of the cancer,indicating that P73 may be a tumor suppressor gene of this malignancy.
